Abstract: | 本實驗分兩大部分:多醣體培養實驗及小白鼠體內抗氧化活性測試。 多醣體培養實驗探討裂褶菌深層醱酵培養多醣之濃度曲線並和傳統的靈芝多醣及最近備受注目的巴西蘑菇多醣做比較,希望能找出裂褶菌多醣的最適收成期間、最好的培養基及最佳培養方式。 小白鼠體內抗氧化活性測試是以培養好的裂褶菌多醣體以口服方式給予小白鼠,探討不同多醣濃度及不同使用期間對小白鼠抗氧化活性的影響。 在多醣的液態深層培養方面,裂褶菌比其他菌種容易生長,產生的多醣量也較多,通氣培養及適當攪拌培養液對多醣的產量都有幫助,而較適合裂褶菌多醣生產的為含3?4%葡萄糖、0.5%酵母抽出物,0.1% KH2PO4及0.05 % MgSO4的培養液,另外裂褶菌多醣經高溫高壓滅菌後,多醣仍存在,且多醣總醣量和滅菌前沒有差異。 在多醣對小白鼠的抗氧化活性測試方面,由肝臟及脾臟的SOD濃度變化趨勢都可看出多醣體的確可以使小白鼠體內的SOD濃度升高而達到抗氧化的效果。 This study was divided into two parts: The first part was to study the in vitro culture of fungus to produce glucan. In this part we tried to use the different ways to culture three kinds of fungi included Schizophyllum commune, Ganoderma lucidum and Agaricus blazei. In fermentative culture, Schizophyllum commune was better than the other two fungi in growth rate and the rate of glucan production. The optimum cultural medium of the Schizophyllum commune included 3~4% of glucose, 0.5% of yeast extract , 0.1% of KH2PO4 and 0.05% of MgSO4, and optimum stirring and aeration will increase glucan production in liquid culture. There was no change when the schizophyllan were treated with high temperature and high pressure. In the second part of this study we wanted to know the influence of SOD concentration when the mice were fed with glucan in different concentration and different feeding days. We finded when schizophyllan were added into drinking water, the SOD concentration in liver and spleen of mice will increase . |
