| Abstract: | 目前對癌症的研究，已知癌細胞的形成和oncogene有密切的關係，本實驗以B104-1 cell為model利用Antisense RNA techingue去抑制癌細胞的oncogene功能，以期瞭解是否可將癌化細胞轉變為正常的纖維細胞（NIH3T3) ．進一步更以Native癌細胞作為觀察對象，是否也能經由抑制其oncogene的功能而轉化為正常細胞． 我們先construct antisense DNA vector，再將antisense vector送入癌細胞，挑選其transfectonts．由southern blot analysis知道Antisense DNA已插入癌細胞的Genomic DNA上．將挑選到的transfectants 做in vitro Biologioal assay(Soft agar assay) ，發現有partially revert現象，另外又發現一種類似revertant cell line．進一步以Native cancer cell做soft agar assay，也發現有partially revert甚至有些transfectant有fully revert回normal cell的現象． |
