dc.description.abstract目前對癌症的研究,已知癌細胞的形成和oncogene有密切的關係,本實驗以B104-1 cell為model利用Antisense RNA techingue去抑制癌細胞的oncogene功能,以期瞭解是否可將癌化細胞轉變為正常的纖維細胞(NIH3T3) .進一步更以Native癌細胞作為觀察對象,是否也能經由抑制其oncogene的功能而轉化為正常細胞.
我們先construct antisense DNA vector,再將antisense vector送入癌細胞,挑選其transfectonts.由southern blot analysis知道Antisense DNA已插入癌細胞的Genomic DNA上.將挑選到的transfectants 做in vitro Biologioal assay(Soft agar assay) ,發現有partially revert現象,另外又發現一種類似revertant cell line.進一步以Native cancer cell做soft agar assay,也發現有partially revert甚至有些transfectant有fully revert回normal cell的現象.
