摘要: | 本研究乃探討帶鱗單角革單棘魨魚皮以納豆菌發酵產物的抗氧化能力與其對皮膚纖維母細胞的影響。結果顯示,納豆菌能有效利用帶鱗單角革單棘魨魚皮為培養基質,並產生蛋白質水解物。有關DPPH、ABT+、超氧陰離子、過氧化氫與氫氧自由基的清除能力,均隨著發酵時間而提昇,而發酵時間延長對於清除過氧化氫的能力最有助益。對於還原力與酪氨酸酶活性的抑制效果則以發酵72小時較佳,且發酵72小時後的樣品能促進皮膚纖維母細胞超氧岐化酶酵素的活性,其能力可能來自於魚皮蛋白質水解物。在促進皮膚纖維母細胞增生方面,以發酵24小時最佳,在500μg/mL濃度下可提升約30%的細胞數。各發酵樣品皆能促進皮膚纖維母細胞原膠原蛋白合成,而發酵72小時促進效果最佳。然而,若先以UVA照射皮膚纖維母細胞,使其在受損的狀況下,未發酵之產物對細胞之修復能力有最佳之功效。但是,發酵24與48小時樣品亦兼具有促進受損之皮膚纖維母細胞存活與原膠原蛋白生成之功能。因此,利用納豆菌發酵帶鱗單角革單棘魨魚皮後,確實可得到具有皮膚保健功能的產物,並且在不同的發酵時間下得到的產物,可能可以分別用在皮膚美白與抗老化的保養品中。 The objective of this study was to produce functional products for skin care by fermenting the skin of Aluterus monoceros by Bacillus subtilis natto. The fermentation products were evaluated for their antioxidant properties and effect on procollagen synthesis of skin fibroblast. The results showed a dramatic increase in scavenging abilities of DPPH, ABT+, superoxide anion, hydrogen peroxide and hydroxyl free radical of the fermentation products. Increase of fermentation time had most significant effect on hydrogen peroxide scavenging ability as compared to other free radicals. As for the reducing power and tyrosinase inhibition activity, the 72h fermentation product was most effective. The 72h product also improved the superoxide dismutase activity of the skin fibroblast. A 30% increase in skin fibroblast proliferation was observed on treatment with 24 hrs fermentation product at a concentration of 500μg/ml. While, fermentation product from 72 hr exhibited maximum procollagen synthesis by skin fibroblast. On irradiation with UVA, skin fibroblasts treated with un-fermented fish skin extract had the most profound effect on the repairing of the UVA-damaged cells. However, the 24 h- and 48 h- fermented products also showed an improved cell viability and procollagen synthesis then untreated cells. Hence, fermentation of the skin of Aluterus monoceros by B. subtilis natto could produce functional products of skin care, and the products with different fermentation times can be used in skin whitening and anti-ageing cosmetics. |
