在分子生物學的研究中,基因微陣列晶片是一項具前瞻性的分析技術,其優點在於可以同時準確的偵測數千至數萬點基因的表現量,大幅地縮短篩選有興趣的目標基因所須之時間;然而,在進行傳統的microarray雜合反應,標的物溶液主要是靠著擴散作用來跟探針鍵結,是一項非常耗時的處理步驟。
有鑑於此,在本研究中,使用微聲流效應來加速液體的混合,進而增加雜合的反應效率,縮短雜合所需的時間,並使用此效應來建構一台新型的生物晶片雜合系統。所架設的雜合系統分為三大系統,微聲流系統產生微聲流用來使標的物溶液混和,溫控系統使雜合的溫度維持在42℃,控制系統控制微聲流系統及溫控系統。此系統的主要優點為所使用的儀器簡單,架設容易且架設經費低廉。然而,於校能驗證的相關實驗中,卻不能達到本論文所預期。因此,於提高雜合效率方面仍然需要做進一步的改善。
Abstract
DNA microarray technology is one of the most promising analytical tools in molecular biology. The advantage of DNA microarray is that it allows parallel analysis of gene expression of hundreds to thousands of genes at the same time. Therefore, it can greatly reduce time in analyzing. However, conventional DNA microarray hybridization relies on diffusion of target to surface-bound probes, and thus is a rate-limited process.
In this study, I use cavitation microstreaming to construct a new microarray hybridization station in order to accelerate the mix of target solution、enhance the hybridization efficiency and reduce the hybridization time.
The hybridization station includes cavitation microstreaming system、thermocontrol system and control system. Cavitation microstreaming system induces cavitation microstreaming in order to mixing up the target solution. Thermocontrol system maintains the hybridization temperature in 42℃. Control system control cavitation microstreaming system and thermocontrol system.
This station has many advantages including a simple apparatus, ease of implementation and low cost. However, it didn’t reach my expectancy when testing the hybridization efficiency. So, this station needs further improvement in enhancing the hybridization efficiency.
