Abstract: | 印度梨型孢真菌 (Piriformospora indica, P. indica) 能與水稻根部共生,並增進植株生長勢及耐逆境能力。本研究以水稻品種臺中在來一號 (Oryza sativa L. cv. Taichung Native 1, TCN1) 為材料,探討P. indica與其根部共生對於提升水稻對瘤野螟 (Cnaphalocrocis medinalis (Guenée), leaffolder) 危害耐受能力之效果,並進一步了解其耐受機制。研究結果顯示,接種P. indica之水稻幼苗在受到瘤野螟幼蟲侵食後之生長勢,比未接種P. indica之植株佳。以接種P. indica水稻幼苗餵食瘤野螟幼蟲,會造成幼蟲體重下降,且化蛹成功比例降低,顯示P. indica除了提升水稻對瘤野螟之耐受能力外,亦會影響瘤野螟幼蟲之生長。水稻植株葉片之基因表現分析結果顯示,OsJAmyb、茉莉酸生合成酵素基因 ALLENE OXIDE SYNTHASE 1 (OsAOS1)、jasmonyl-amino acid synthetase 基因 JASMONATE RESISTANT 1 (OsJAR1)及JA–isoleucine (JA–Ile) 接受器基因CORONATINE INSENSITIVE1 (OsCOI1) 之表現量受P. indica共生而有所提升,推測P. indica 可提高水稻內生茉莉酸含量並增強茉莉酸相關之抗蟲訊息傳導。此外,藉由植株體內會影響害蟲生長之胰蛋白酶抑制劑(trypsin proteinase inhibitor, TI)、苯丙胺酸裂解酶 (phenylalanine ammonia-lyase, PAL) 及多酚氧化酶 (polyphenol oxidase, PPO) 活性分析,發現接種P. indica之植株受到瘤野螟幼蟲危害後有較高之上述抗蟲物質活性表現。根據上述實驗結果,P. indica可能是透過增強植物茉莉酸相關之防禦途徑,並增進抗蟲物質累積,以提升水稻對瘤野螟危害之耐受能力。 The mutualistic symbiosis between Piriformospora indica (P. indica) and rice roots resulted in promotion of plant growth and increase of stress tolerance. Here, we focused on studying the effect of P. indica on rice (Oryza sativa L. cv. Taichung native 1, TCN1) tolerance against Cnaphalocrocis medinalis Guenée, (leaffolder) and the related P. indica induced herbivore defense mechanisms. The results showed P. indica-colonized rice had higher biomass and photosynthetic activity than non-colonized rice under leaffolder infestation. Leaffolder larvae fed with P. indica-colonized rice had lower relative growth rate and pupal ratio than that with non-colonized plants. This indicated P. indica can enhance rice tolerance against leaffolder and also present negative effects on larvae growth. According to gene expression analysis, in leaves of leaffolder infestated rice plants expressions of jasmonate (JA)-related genes such as ALLENE OXIDE SYNTHASE 1 (OsAOS1, JA biosynthesis gene), JASMONATE RESISTANT 1 (OsJAR1, jasmonyl-amino acid synthetic gene), CORONATINE INSENSITIVE1 (OsCOI1, JA–isoleucine (JA-Ile) receptor gene) and OsJAmyb in leaf tissues of P. indica-colonized plants were higher than those were in non-colonized plants under leaffolder-infestated conditions.It was suggested that P. indica enhance leaffolder tolerance of rice plants may be mediated the activation of JA-related signaling pathway. Moreover, P. indica also enhanced the activities of defense-related proteins, e.g. trypsin proteinase inhibitor (TI), phenylalanine ammonia-lyase (PAL) and polyphenol oxidase (PPO). Taken together, the symbiotic interaction between P. indica and rice lead to enhance JA defense pathway and several defense-related protein activities to reduce the leaffolder caused damage. |
