Abstract: | 本研究利用81個RFLP標誌,包括55個縐葉菸草基因組選殖體、13個縐葉菸草cDNA庫選殖體、5個栽培菸草cDNA庫選殖體、3個縐葉菸草酵母菌人工染色體末端、一個26S rDNA和一個硝酸鹽還原酵素基因建立縐葉菸草之遺傳圖譜。所偵測得到在縐葉菸草和長花菸草F2族群中分離情形的資料,初步建構縐葉菸草10個連鎖群,共964.9 cM。根據本研究室楊人泰同學利用酵母菌人工染色體末端B172及王中茹同學以26S rDNA螢光原位雜交的結果,推測含B172標誌的第一連鎖群屬於縐葉菸草的第一條染色體,而含26S rDNA的第十連鎖群屬於縐葉菸草的第九或第十條染色體。 A total of 81 RFLP markers, including 55 Nicotiana plumbaginifolia genomic clones, 13 N. plumbaginifolia cDNA clones, 5 N. tabacum cDNA clones, 3 N. plumbaginifolia YAC end clones, a 26S rDNA and a nitrate reductase gene, were used to construct a linkage map for N. plumbaginifolia in this study. Inheritance of the markers in F2 population of N. plumbaginifolia × N. longiflora was analyzed. A preliminary genetic map containing 10 linkage groups covering 964.9 cM was established. According to the results from fluorescence in situ hybridization using B172 YAC clone (Yang, 1998) and 26S rDNA (Wang, 1998) as probes, we suggest that linkage group I belongs to chromosome 1 and linkage group X belongs to chromosome 9 or 10 of N. plumbaginifolia. |
