Abstract: | 在比較每個細胞所含 G6PD 之活性(International unit ; I. U .)我們發現人類淋巴細胞之含量為紅血球的 116 倍,且人類淋巴細胞之 G6PD 對 G6PD (Glucose-6-phosphate)及NADP+之動力常數 (kinetic constant;Km)有相當大的差異. 我們在人類淋巴細胞的初級培養(primary culture)時發現, G6PD 在單位重量蛋白質中之活性與細胞之成長複製有正相關性.且發現在 FBS (Fetal Bovine Serum)中存在一個 G6PD 的抑制劑 ( inhibitor ) ,它既抑制 G6PD 之活性,亦抑制細胞之成長.我們也發現,在透析過血清(dialyzed FBS)的培養中,加入核糖核?(ribonucleosides)及去氧核糖核?(deoxyribonucleosides),能夠幫助突破IL-2(Intexleukin-2)對培養的淋巴細胞所造成刺激其生長的限制,而 G6PD 亦呈現持續增強的趨勢. The G6PD activity varied with different human blood donors. In average, the enzymetic activity of human lymphocyte was about 116 times that of erythrocy-te. The Michaelis-Menten constant of human lymphocyte G6PD for either glucose-6-phosphate or NADP+ was variable among individual blood donors. In the primary culture of human lymphocyte, the G6PD activity increased as the cultured cells were proliferous. We found that there might exist some factor(s), which was dialyzed away from fetal bovine serum, suppressed not only the G6PD activity but also the cell growth. As cells were cultured in either fetal bovine serum or the dialyzed serum, the cell growth could be stimulated by the addition of Interleukin-2. Meanwhile, addition of ribonuc1eosides or deoxyribonucleosides enhanced the G6PD activity and the cell growth. |
