摘要: | 台農67號種子經成熟胚誘導的癒合組織,分別培養於含tryptophan或tryptophan加benzyladenine的MS培養基中,其植株生率很低,不及5%,而培養於含1ppm之naphthaleneacetic acid及2ppmkinetin者,其再生率可高達26%。 電泳分析圖顯示,癒合組織及懸浮培養細胞,皆具有3條酸性磷酸?的異構?帶,根部則具有7條,而葉卻只有6條,再者於以上各組織同時出現有一條共同的低分子擴散帶;另外以DEAE管柱純化乳熟期種子,發現有5個酸性磷酸?的活性高峰,再以電泳分析,同樣具有5條異構?帶。 經Gomori溶液處理,藉電子顯微鏡觀察酸性磷酸?的次細胞分佈,癒合組織中酸性磷酸?分佈於細胞壁,細胞核及液胞中,又於分化培養基14天已綠化之組織,除了前提各胞器外,於綠化期間,大量含有澱粉粒的色素體內,也具有高活性的酸性磷酸?,和癒合組織比較,在葉中,酸性磷酸?也出現於粒線體,高爾基及細胞質中,而根部於分生組織細胞內,分佈較廣,如細胞核,細胞壁,粒線體等,在延長部的細胞則主要出現於細胞壁內。 Calli, induced from mature seed of rice (Oryza sativa L. cv. Tainung No. 67), were cultured in MS medium containing tryptophan or tryptophan and benzyladenine. Their frequencies of plant regeneration were very low, only 5%. However, the MS medium supplemented with 1 PPm naphthaleneacetic acid and 2 ppm kinetin could improve the frequency of plant regeneration up to 26%. Electrophoretically analyzing isozymes of acid phosphatase in different plant tissues or organs, there were three bands in calli and suspension cultured cells, seven in root, six in leaf, and five in milky seed. Subcellular localization of acid phosphatases by the method of Gomori showed that they mainly distributed in cell wall, nucleus and vacoule in callus tissue. In greening calli ready to differentiation, acid phosphatases were also found in plastid. In leaf, they were also present in mitochondria, Golgi and cytoplasm. While in root except meristematic tissue, acid phosphatases mainly appared in cell walll. |
