摘要: | 本研究乃以在光照下生長八天的台南十一號玉米幼苗第一本葉的切離葉片段為材料,探討在黑暗誘導老化期間,苯甲腺嘌吟(N6-benzyladenine)延緩葉片老化的可能機制。 實驗結果顯示,葉綠素、蛋白質、核糖核酸含量均隨葉片黑暗誘導老化的天數增加而減少。於葉片切離第六天,對照組這三種含量分別降至為初值的17%、21%、48%;處理組則分別仍維持在初值的80%、79%、69%。苯甲腺嘌吟有顯著抑制葉片老化的效果。於切離葉片老化期間,以血紅蛋白為受質所測到的蛋白質分解?及核糖核酸分解?的活性皆增加。 玉米切離葉片以(上標 3)^H-leucne標記3小時,經過不同黑暗誘導老化期間,測其3H-leucine併入蛋白質量,發現處理組與對照組的(上標 3)^H-leucine 併入量皆減少,且在相同黑暗處理時間下,處理組的(上標 3)^H-leucine併入量較對照組的約高2倍,另外,抽取其總蛋白質,在自動放射顯像圖上,發現在葉片老化期間,處理組的總蛋白質類型與零天(剛切離葉片)的相同。對照組的總蛋白質類型在黑暗處理前2天,與零天的相同,而自第四天起,相應於零天的六條主要蛋白質帶,分子量為80,000,69,000,62,000,52,000,30,000,及26,000 daltons等,逐漸減少或消失。 由玉米切離葉片總蛋白質及葉綠體蛋白質類型的電泳分析,發現處理組於葉片切離六天後,其蛋白質類型仍與零天的相同。對照組在黑暗處理前2天仍與零天的相同,但自第四天起,相應於零天的主要蛋白質帶逐漸減少或消失。在葉片總蛋白質與葉錄體蛋白質類型均減少或消失的主要蛋白質帶有八條。 在黑暗下經過六天的切離葉片葉綠體在試管中蛋白質合成能力分析,指出處理組的葉片葉綠體在試管中蛋白質合成能力較對照組高很多。 The detached primary leaf segments from eight-day-old illuminated corn seedlings (Zea mays L. cv. Tainan No. ll) were used to study the effect of N6-benzyladenine (BA) on the retardation of senescence in the dark. A decline of chlorophyll, protein and RNA mark the progress of senescence in detached leaves. BA at the concentration of 1×10^-5M was effective in retarding the decline in the amount of chlorophyll, protein and RNA. The activities of hemoglobin-degesting protease and ribonuclease in detached leaves increased during senescence. Leaf segments were prelabeled with 3^H-leucine for 3 hours, and then transferred to medium containing unlabeled leucine with or without BA. The radioactivity of 3^H-proteins in BA-treated leaf segments was two-fold higher than in control. The protein patterns in each tissue were analyzed by 12.5% SDS polyacrylamide gel electrophoresis and followed by autoradiography. From the results showed that the total protein patterns for control and BA-treated leaf segments were identical with initial. The molecular weight of major protein bands were 80,000, 69,000, 62,000, 52,000, 30,000, and 26,000 daltons. But these major protein in control began to decrease or disapprear on 4th-day during senescence. From SDS-polyacrylamide gel electrophoresis of total Proteins and chloroplast proteins, showed that the protein paterns for control and BA-treated leaf segments were identical. In both kinds protein patterns in control, there are eight main protein bands in common began to decrease or disappear on 4th-day during senescence. The in vitro protein synthesis activity of chloroplasts isolated from detached leaves after incubation in the dark for 6 days, showed the chloroplasts of BA-treated detached leaves have much higher biosynthetic activity than control. |
