dc.description.abstract | Saccharomyces cerevisiae被廣泛的運用在自然界中,且與日常生活中息息相關,其發酵過後產生大量之副產物,而此副產物含有豐富的蛋白質、多醣類和維生素,並且被認定為是安全無疑的。S. cerevisiae的細胞壁主要成分有β-glucan, chitin和mannoprotein,有研究指出細胞壁多醣展現了多樣性的生物活性效果,例如:抗腫瘤、抗癌、抗病毒、抗凝集以及具有免疫反應的產生。4-Nitroquinoline-1-oxide是廣泛被使用的致突變劑和致癌劑,經由代謝活化後具有致癌性,此化學致突變劑常運用在腸道系統的研究。本實驗探討從三株S. cerevisiae萃取出來的細胞壁多醣,處理對於4NQO誘導intestine 407細胞DNA傷害之抗細胞毒性研究及抗細胞毒性效應,分別藉由去致細胞毒性效應、阻斷效應和生物抗細胞毒性效應,並利用彗星電泳實驗測試,期望細胞壁多醣可以減低4NQO對於Int-407細胞之基因毒性。結果顯示三株S. cerevisiae細胞壁多醣分別在5、10、20 μg/mL濃度對於只有給予4NQO這組有顯著性的差異並且提升了細胞存活率達85%。在探討抗細胞毒性效應時,細胞壁多醣對於抑制4NQO誘導人類正常腸道上皮細胞Int-407之細胞毒性,推測是經由阻斷效應和生物抗細胞毒性效應作為保護細胞之效應,能調整細胞機能來抵抗4NQO的致突變作用,並幫助細胞修復損傷。在抑制基因毒性方面,S. cerevisiae細胞壁多醣也能降低4NQO誘導Int-407產生的基因損傷,損傷分數由164.00 – 169.87降至19.00 - 33.01,顯示S. cerevisiae細胞壁多醣確實能使人類正常腸道上皮細胞Int-407降低4NQO造成的基因毒性傷害。 | zh_TW |
