Abstract: | 本試驗在探討光質與光強度對草莓組培苗再生的影響,試驗材料為`桃園一號'和「長柄」兩品種,以MS培養基為基礎培養基,培養於微型組織培養系統(Mini-E Light System, MEL,光茵生物科技公司,臺灣)。以MS培養基進行走莖之莖頂組織的誘導,在光強度45μmol∙m-2∙s-1下誘導4週,結果顯示光質組合對兩品種的莖頂組織誘導率皆無顯著影響;但品種間差異顯著,`桃園一號'的誘導率介於20% - 40%,而「長柄」誘導率介於50% - 90%。另以添加BA 2mg∙L-1的MS培養基進行芽體增生的試驗,先以6種光強度54、45、34、25、20、7 μmol∙m-2∙s-1進行初步的篩選,針對芽體數和葉片數的增加量,以及芽體鮮重的增加之效果,篩去表現不佳的條件,結果以光強度45、34和20 μmol∙m-2∙s-1進行後續芽體增生階段之光質組合的試驗。以3種光強度和8種光質組合培養4週後,無論是在芽體增生、葉片數和鮮重的增加量,對兩個品種`桃園一號'和「長柄」來說,光強度34 μmol∙m-2∙s-1和光質組合7R1G1B均為芽體增生階段的最佳條件,而兩品種間具有明顯差異,「長柄」的表現明顯優於`桃園一號',平均每個培殖體增加了12.33個芽體、3.41個葉片、鮮重0.67g。最後,以MS培養基進行組培苗的發根,兩品種間發根率無顯著差異,`桃園一號'和「長柄」的發根率平均都高於90%。綜合本試驗結果,建議草莓再生的光強度及光質組合為34 μmol∙m-2∙s-1和7R1G1B。 The aim of this research is to study the effect of light intensity and light quality on micropropagation of Strawberry ‘Taoyuan No. 1’ and ‘Changbin’. Shoot-tip meristem was cultured in MS medium at a Mini-E Light System for shoot regeneration under 45 μmol∙m-2∙s-1 for 4 weeks, the regeneration rate was significant different between cultivras but not among 8 light quality combinations (9CW, 9WW, 9B, 9R, 8R1B, 7R1G1B, 7R1B1IR, and 3R3B3IR). The regeneration rate of ‘Taoyuan No. 1’ and ‘Changbin’ is ranging between 20% - 40% and 50% - 90%, respectively. And then following the experiment of shoot proliferation in MS medium with BA 2 mg∙L-1 , light intensity of 45、34 and 20 μmol∙m-2∙s-1 were selected from a screening of 54、45、34、25、20、7 μmol∙m-2∙s-1. After 4 weeks of shoot proliferation, both cultivras obtained highest shoots, leaves and fresh weight as cultured under 34 μmol∙m-2∙s-1 of 7R1G1B. Besides, the proliferation was significantly higher in ‘Changbin’. Changbin'increased 12.33 shoots、3.41 leaves and fresh weight of 0.67g per explant in average. For rooting in MS medium, more than 90% of the explant successfully emerged roots, and the rooting was not significantly affected by cultural condition including light intensity and quality combinations. According to the result, mixed light of 7R1G1B at 34 μmol∙m-2∙s-1 was suggested strawberry micropropagation including shoot regeneration, proliferation and rooting. |
