摘要: | 尋找有效且經濟的方法清除環境污染一直是科學家努力的目標。利用真菌分佈廣泛,繁殖迅速的特性,篩選其中抗鎘的品種就成了一種可行的生物救濟法(bioremediation)。本實驗比較Curvularia lunata KYC 9604以及自菌種中心所購得兩株不同品系的菌株,在不同鎘濃度(0ppm,100ppm,500ppm,1000ppm)處理下,由其菌落半徑及菌絲乾重變化情形,觀察品系間抗鎘性狀及反應的不同。 實驗結果顯示,KYC 9604相較於其他兩品系,對鎘具有較好的抗性,對環境中鎘也具有快速的吸附能力。以100ppm的鎘處理15分鐘後菌絲的吸附量達到69ppm。超微構造的觀察顯示,鎘處理後細胞的油滴與液胞數目增加,而細胞壁外側的黑色素(melanins)則在鎘處理後消失,細胞內的油滴並有向細胞壁移動進行胞外分泌(exocytosis)的情形。比較利用X光能量分散成份分析儀(EDS),偵測不同菌絲乾燥方法的樣品中鎘含量變化情形,以利用冷凍乾燥法處理的樣品偵測效果最好,偵測所得樣品中鎘的重量百分比變化,相較於原子吸光光譜儀所測得鎘含量,兩者增加的趨勢大致相同。最後利用蛋白質電泳,觀察細胞內蛋白質變化,結果有數個蛋白質帶的強度隨處理時間而有所不同。 Scientists are always involved in removing heavy metal pollution from environment in effective and ecnomic methods. Using the wide distrubition and fast regenerating characteristics of fungi, screening cadmium resistant strains is a feasible bioremediation. In this study Curvularia lunata KYC 9604 and two different strains bought from culture center were investigated. The different in the cadmium resistant trait and response among these strains was studied from their alternation in colony radius and dry weight for different cadmium concentration (0 ppm, 100 ppm, 500 ppm and 1000 ppm). KYC 9604 has better cadmium resistance, and fast cadmium adsorbent ability than the other two strains. After 15 mins in 100 ppm cadmium treatment, cadmium uptake by myclium was up to 64 ppm. Oil drops and vacuoles were increase after cadmium traet, and the melanins outside the cell wall were disapper. Exocytosis occured when vacuoles were move toward the cell wall. Cadmium content in different smaples were detected by EDS. Compare these sample drying methods, the result using freeze drying was the best. After all, we use SDS-PAGE to view the protein patterns alter, some protein patterns were also altered by the duration of cadmium treatment. |
