摘要: | 以丁基拉草對水稗種子作萌芽前處理,觀察種子萌芽,及幼苗生長的變化。丁基拉草不抑制水稗種子萌芽,但是延後萌芽的時間;對幼苗的生長則有明顯的抑制。在形態方面,處理組的幼苗發生兩大變化:(1)第一片葉子不突出子葉鞘,(2)根的生長被抑制。由解剖切片觀察幼苗根尖的細胞,處理組的細胞發生囊泡化(vesiculation)現象,逐漸擴大的液泡將細胞質嵌入(invagination),最後細胞趨向崩解。在生化方面,處理組的幼苗在澱粉分解酵素的活性受到50%的抑制,胚乳碳水化合物的分解也被抑制;在蛋白質分解酵素則隨不同受質而有不同的變化,以牛乳蛋白為受質時,酵素活性被抑制,同時胚乳蛋由質的分解也被抑制,而以BAPA為受質時,酵素活性不被抑制,同時處理組的幼苗的可溶性胺基酸含量逐漸增加,顯示丁基拉草使水稗幼苗細胞發生自我分解(autolysis);在過氧化酵素,則活性被抑制,而異構?的型態沒有明顯的變化。 水稗種子在丁基拉草溶液中播種36小時,抽取其中核糖體,以連續蔗糖濃度梯度法分離的結果顯示丁基拉草在水稗萌芽早期就抑制形成polysome的能力。 水稗種子在不同濃度的丁基拉草溶液中播種12小時,然後經不同時間的水沖洗,再移植到水中生長,觀察其幼苗的形態,顯示即使以低濃度(6ppm)的丁基拉草處理,經過較長時間的沖洗也不能除去丁基拉草的抑制作用,只能恢復一部分生長。可能丁基拉草對水稗萌芽早期就有相當的抑制作用。由以上的觀察推測丁基拉草對水稗的作用機制是多方面的,抑制蛋白質合成是其中之一。 The effects of butachlor [2-chloro-2’, 6’-diethyl-N-(butoxymethyl) acetanilide] on seed germination of barnyardgrass (Echinochloa crusgalli Beauv.) and subsequent seedlying growth were investigated. Butachloi did not affect seed germination up to a concentration of 100 ppm tested in this study, but delayed the time of coleorhiza emergence through the pericarp of seed for 12 hours. Further seedling growth was severly inhibited. Emergence of the primary leaf from the coleoptile and root growth were inhibited by butachlor as low concentration as 6 ppm. Light and electron microscopic studies indicated that butachlor induced cellular autophagy of root tissues. The development of amylase and protease activities were prevented so that the degradation of reserve carbohydrates and proteins in endosperm was affected by butachlor during 7 days of germination. Protein synthesis namely polysome formation was also inhibited during the first 36 hours of germination. Time course studies on the seedling growth revealed that the inhibition was observed during the initial 12 hours of seed imbibition. |
