dc.description.abstract | 本研究中利用採自宜蘭五結鄉健康養殖草蝦肝胰臟的Vibrio anguillarum UAH-3.以各種不同的生理環境,包括不同鹽度、PH值溫度及營養度等來培養,並進行其細胞外蛋白?的純化及各項形質分析,結果發現分別在鹽度30?、pH9、溫度20℃時供試菌株產生之蛋白?具有最高的活性。細胞外產物經Sephadex G-200 column分離得到兩種蛋白?,其分子量分別為82k及 64k。經測試各項性徵實驗,發現其最適合作用的溫度為 45℃,最適合的 PH 值為7,經60℃加熱10分鐘即失去活性。在抑制劑( inhibitor )的測試中,1,10-phenaethroline及 ethylenediamine tetraacetic acid(EDTA)對其有抑制作用,phenylmethanesulphonyl fluoride( PMSF ),pefoloc SC, pepstatinA及leupeptin則否,証實為metalloprotease,且其蛋白?活性與 Ca 2+及Mg2+有關。 純化後的蛋白?對草蝦的毒性試驗結果,會造成注射部位的肌肉壞死,在顯微鏡下肌肉組織有明顯的壞死灶,周圍並出現血淋細胞浸潤及結締組織增生等現象,隨著接種時間的延長,炎症反應的範圍有擴大的趨勢。 | zh_TW |
