摘要: | SCD2 (Stearoyl CoA desaturase2)的負調節序列PRE(Preadipocyte repressor element)的序列和一普遍存在之核內因數YY-1 的結合序列甚為相似。YY-l對轉錄的調控有時扮演促進子的角色,有時則扮演抑制子的角色,端視所涉及的基因以及結合序列而定。
在本論文中,我經由比較UCR(upstream conserved region)結合序列CGCCATTTT與SCD2?動子PRE結合序列AGCCATTTC。發現一個共同因數可以結合到這二者的結合序列上,從競爭性的原性電泳膠遲緩試驗中,我已經證實 UCR和SCD2的序列專一性的確很相似。更進一步研究顯示UCR和SCD2上的結合因數具有相同抗原性,於是,我們可以下一個結論:普遍存在之核內因數,YY-1,可以認知SCD2?動子上的PRE。 至於YY-1在SCD2上扮演何種角色,尚需進一步實驗才得以知曉。 The sequence of a negative cis-element, PRE (preadipocyte repressor element) of SCD2 (stearoyl CoA desaturase 2) is closely related to the binding sites for a ubiquitous cellular factor, YY-1 (NF-E1,δor UCRBP). YY-1 appears to be somewhat unique in that it can exert either a positive or a negative control over transcription, depending on the genes and binding motifs involved. In this thesis, I compared UCR (upstream conserved region) core sequence CGCCATTTT with SCD 2 promoter binding site PRE AGCCATTTC. I have detected common factors that can bind to these two motifs. From EMSA competition assay, I have confirmed that the sequence specifity of UCR and SCD 2 binding factors are similar to YY-1. Further studies have shown that both UCR and SCD 2 binding factors share antigenicity with YY-1.We conclude that the ubiquitious transcriptional factor ,YY-1 ,can recognize PRE of SCD 2. Further experiments are needed to clarify the functional role of YY-1 on the regulation of SCD 2 expression. |
