Title: Candida cylindracea 所生產的 lipase OF 之基礎研究

Abstract: 脂肪酵素是工業上常用的一種酵素。由酵母茵 Candida cylindracea所生產的脂肪酵素 OF ,經簡單純化後,可得到很純的蛋白質,做 N 端的胺基酸序列分析可得到將近四十幾個胺基酸序列。將這一段 N 端序列的兩端,翻譯成遺傳密碼後,利用 DNA 自動合成儀合成兩端引發物(primer)。再利用細胞 DNA ( genomic DNA )為摸板,以 Polymerase Chain Reaction (以下簡稱 PCR )的方法,定出這一段一共九十二個鹼基( base pair )的核酸序列。接著以這一段核酸序列當探針,利用南方墨漬法(southern blotting ) ,釣出一段約9kb的DNA片段,經過基因選殖( cloning )後,這段 DNA 發生了刪除及重組,只剩下約1.3kb ,以核酸定序法定出這段 DNA 的核酸序列後,無法對出探針位置的所在。由於這個基因不穩定,在基因選殖過程中,一再發生刪除及重組的現象,最後選擇建立一個 cosmid genomic library ,挑選出兩個完整的菌落(clone),並以探針找出 lipase OF 基因所在位置。
除了分子生物方面的研究外,本篇論文並探討脂肪酵素 OF 對基質的特異性,包括它的定位選擇性及鏡像選擇性。脂肪酵素 OF 對卵磷脂第一位置酯鍵及第二位置酯鍵並沒有很高的選擇性,而在鏡像選擇性方面,由於脂肪酵素具有兩個基質結合位置,一個是對酸類,一個是對醇類,透過酯化反應,可以一次對消旋酸及消旋醇同時作解析(resolution )。利用消旋2- phenoxypropionate 衍生物及一些消旋醇當反應基質,藉由脂肪酵素 OF 的作用,可以對這些消旋化合物作部分解析。
Lipase OF from Candida cylindracea is very useful enzyme in the industry . After the purification , the N-terminal of the lipase has been sequenced about forty amino acids . Using the genomic DNA as the template of Polymerase Chain Reaction , and the synthesis of two primers deduced from the known N-terminal sequence , the DNA sequence corresponding to the N-terminal of the lipase was determined . This sequence was used as the probe to find the gene of Lipase OF from genomic DNA . A clone was sequenced , but this sequence is not consistent with the N-terminal sequence of Lipase OF , probably due to the deletion and gene recombination in the cloning experiment . Besides the respect of molecular biology , the substrate specificities of Lipase OF were also studied . Lipase OF doesn’t have good regioselectivities on phospholipid substrates , but Lipase OF can resolve partially both racemic acids and racemic alcohols in the esterification reaction in organic solvent simultaneously .
