dc.description.abstract | 促性腺激素(gonadotropin)有促進魚類排卵(ovulation)以至產卵(oviposition)的功能。本研究以離體(in vitro)試驗的方法探討在最後成熟(final maturation)過程中各主要時期,卵核偏中央(subperipheral )期、卵核移動前期(the early stage of germinal vesicle migration )、卵核移動後期( the late stage of germinal vesicle migration )、卵核破裂( germinal vesicle breakdown , GVBD)期的泥鰍(Misgurnus anguillicaudatus)卵細胞(oocyte ) 對人類絨毛膜促性腺激素( human chorionic gonadotropin , HCG )不同劑量: 0.01 、0.1 、0.5 、1、5μg/m1 的刺激反應。以卵細胞內每單位蛋白質環腺核甘單磷酸(cyclic AMP)的累積量作為反應程度的指標。目的在藉以瞭解 HCG 對於誘導及控制卵細胞最後成熟的機制,以期作為人工促進產卵技術之建立及應用的依據。自牛腎上腺抽取分離所得的結合蛋白質(binding protein)其專一性(specificity)足以精確測定 cyclic AMP 之含量。環腺核甘單磷酸含量的測定乃是利用競爭性蛋白質結合原理(the principle of competitive protein binding )為基礎的環腺核甘單磷酸結合蛋白分析法(cyclic AMP binding protein assay )。蛋白質的測定則用 Lowry method 。為了精確測定未知濃度的 cyclic AMP ,建立可靠之分析方法,對 cyclic AMP 結合蛋白質的特性進行一連串的實驗。其結果為結合蛋白質對 cyclic AMP 有專一性的結合,且有高親和常數(affinity constant ) 其值為 3.22×l0-9 及低結合容量(binding capacity )其值為 2.71 picomole /mg protein 。環腺核甘單磷酸結合蛋白分析法的最適條件為培養溫度4℃;培養時間三小時;氚標誌環腺核甘單磷酸(3H-cAMP)含量為5nCi ;結合蛋白質由原液稀釋到可達32.9%結合度的濃度(本實驗為1.14mg protein/m1); cyclic AMP 含量的測定範圍在0.25 pmole 與 10pmole之間(結合度 81.18-23.17% )。在卵細胞最後成熟過程中,泥鰍卵細胞對於HCG的離體作用,僅在卵核偏中央期有明顯的反應,即隨著 HCG 濃度的增加,每單位蛋白質 cyclic AMP 的累積量有增加的趨勢。至於卵核移動前期及後期和卵核破裂期的卵細胞對HCG 的反應有消失的現象。就生理調控機制的觀點,其可能的原因有(一)促性腺激素(HCG)對於促性腺激素接受器(HCG receptor)的抑制調節(down regulation)或(二)促性腺激素接受器在處理前已達飽和(saturation)。就生理的意義而言,表示促性腺激素控制最後成熟的重要性,已為其他激素(hormone)或類固(steroid)等取代。 | zh_TW |
