dc.description.abstract | 利用親和性管柱,配合DEAE-Sephadex A-50與CM-Sephadex C-50離子交換層析,我們成功地純化出人類紅血球中之六磷酸去氫?(G6PD)。當使用SDS-膠體電泳鑑定其純度時,只看到單一蛋白質色帶,分子量約為55 Kd。不含SDS之非連續電泳膠體中,其活性染色與蛋白質染色之色帶均為單一,且位置相同。由以上結果可判斷其為純質(Homogeneous)。 在含1M Urea, 6% Acrylamide之等電荷焦集電泳(Isoelectrofocusing,IEF)中,純質G6PD亦如同粗質G6PD般有7個同功?,且pI值相同,但在1M Urea之中,均質G6PD活性較低。 利用銀染色測定各同功?蛋白質相對量,再與由活性染色所測得之相對活性相比,我們可以求出其中4個同功?之相對單位質量活性(Relative specific activity)。 再與同功?之動力參數相比較,可知還原NADP+能力越強之同功?,其相對活性越高,且除pI6.59的同功?(Band 1)外,pI值高者之活性高於pI值低者。 | zh_TW |
