Abstract: | 萄葡糖六磷酸去氫?(G6PD: MADP+1-oxidoreductase, E.C.1.1.1.49)在人類紅血球的生理機能中扮演重要的角色,G6PD即在1M urea存在下進行六小時等電荷焦點電泳(isoelectricfocusing IEF)使用Specific activity dye可染出解析度極佳的七個以上G6PD色帶等電點分別為PH6.56, 6.38, 6.21, 6.06, 5.92, 5.80及5.70並經證明大部分為雙體(dimer),故為電荷同功?(Charge isoenzyme)。 將含有isoenzyme之四個色帶切下分別分離濃縮,並測定各isoenzyme之Michaelis-Menten Constant,發現各isoenzyme對Substrate即葡萄糖六磷酸和輔?NADP+之Km和Vmax / Km依各isenzyme之electricfocusing point之位置而改變。 不同來源之新鮮血液和不同年齡的紅血球,其G6PD經IEF分離,皆有相同的七種isoenzyme,但在G6PD的總量中分佈各有不同。 |
