乙型肝炎表面抗原的分離及純化

Abstract

乙型肝炎是一種具有極高傳染性的疾病，以病毒的表面抗原所做成的試劑及疫苗，可望能降低其感染率。在此引用二種方法來純化此表面抗原。它的成分除了蛋白質外，還有一些脂質及醣類，第一個方法便是利用此種特性，將表面抗原從複雜的血清中分離出來，經過第二各步驟的純化，得到15％的回收率。第二個方法是利用抗體的吸附作用，只經單一步驟，便將抗體分離出來，且回收率高達80％，是以非常有效而可行的方法。

Hepatitis B virus infection is a serious public health problem in Taiwan. Hepatitis B surface antigen can be used as reagent and vaccine. The aim of this study was to purify this antigen on large scale. Two methods were studied. One was the affinity chromatography. This was based on the glycolipoprotein nature of the antigen. Through 3 steps purification, 15% yield was obtained. The other one was the immunosorbent. This single step purification was a promising method for our purpose. It gave an 80% recovery.
