| Abstract: | Based upon its ability to inhibit the contractions of electrically stimulated ileum, opioid peptide and its precursor have been isolated from porcine pituitary glands by extraction into acetone, partial fractionation by NaCl, ion-exchange and high performance liquid chromatography. When the component V-2 (with a molecular weight around 14,000) is subjected to trypsinization, its tryptic products cause a profound, shortlasting inhibitory effect on guinea pig ileum. The inhibitory effect can be completely antagonized by naloxone, an antagonist to narcotics. Moreover, this component possesed oxytocic activity on the rat uterus, a target organ of oxytocin. The active fragments for opioid activity are isolated and the amino acid composition is presented. Although additional work is required to confirm their biological activity, components XIII-d and XIII-c have amino acid compositions close to that of ACTH. In this study high performance liquid chromatography has been used for the isolation and purification of proteins and peptides with a wide range of molecular weights. The results are very satisfactory and demonstrate the great potential of this technique in protein chemistry. |
