摘要: | 美洲棉蛉蟲病毒 (Heliothis zea Nudivirus-1, 以下簡稱為HzNV-1) 為一種昆蟲病毒,可同時在多種昆蟲細胞株內引發急性及潛伏性感染。先前在潛伏感染期間發現HzNV-1有一基因persistency-associated gene (pag1) 表現非常多個小分子RNA (miRNAs),並且是唯一一個在潛伏期可被偵測到的HzNV-1基因。前人研究顯示兩個由pag1基因所產生的miRNAs會抑制HzNV-1的早期基因HindIII-I 1 (hhi1),hhi1在宿主體內會導致強烈的細胞凋亡,並且增加急性感染對潛伏性感染的比例。Pag1基因減少hhi1的表現並更進一步地促進潛伏期的建立。許多的報導指出哺乳動物皰疹病毒 (Herpes simplex virus, HSV) 的感染與染色體結構的後基因調控 (epigenetic regulation) 高度相關;然pag1基因本身為一段不轉譯蛋白質的轉錄片段,其可能也藉由自身產生的miRNAs在感染期間調控宿主的染色體結構,但實際上pag1基因如何調控病毒的感染性狀還尚未明瞭。在本實驗中,我們發現pag1基因進入宿主後改變了宿主體內甲基轉移酶su(var)3-9的基因表現量,並發現pag1所產生的Hv-miR-420會抑制su(var)3-9,因此Hv-miR-420這個miRNA可能在HzNV-1感染期間造成宿主的組蛋白修飾改變。此試驗不只提供對HzNV-1感染途徑更清楚的研究,更以不同角度探討病毒miRNAs與宿主後基因調控之間的關係。 Heliothis zea nudivirus-1 (HzNV-1) is an insect virus that can induce both lytic and latent infections in many insect cell lines. During latent infection, the persistency-associated gene 1 (pag1) expresses several microRNAs (miRNAs) and is the only detectable HzNV-1 transcript. Two of the miRNAs derived from pag1 have been found to suppress and silence HindIII-I 1 (hhi1), an early gene of HzNV-1. Hhi1 induces strong apoptosis and increases the lytic to latent ratio. Pag1 reduces the hhi1 expression and further promotes the establishment of latent infections. Previous reports have shown the herpes simplex virus (HSV) is highly associated with epigenetic regulation of the chromatin structure and is regulated upon infection. Since pag1 is a non-coding transcript, it could regulate host chromatin structures through miRNAs upon infection. Nevertheless, the exact pag1 mechanism that alters viral infections is still unknown. In this study, we found that histone modification associated enzymes, su(var)3-9, changed upon pag1 transfection. Pag1 encodes miRNA, Hv-miR-420, to suppress histone methylation transferase, su(var)3-9. As a result, this miRNA causes histone modification in order to HzNV-1 infection. This study not only provides a better understanding of the HzNV-1 infection pathway, but also the relationship between viral miRNAs and epigenetic regulation. |
