摘要: | 蘑菇自從遠古時代就因為其富含營養價值,以及可作為機能性食品和潛在藥物開發的特性而被廣為食用與研究。近年來,由於大眾對健康意識的提升,因此許多研究便著重在在機能性食品的開發。先前的研究顯示,自磨菇中所萃取出的活性分子被認為具有抑菌、抗氧化、抗腫瘤,和免疫調節等特性。 在本篇研究中,選用了Morchella esculenta(羊肚菌)作為實驗材料,並以活性導向的方式進行萃取和分離。首先將破碎後的羊肚菌以有機溶劑(乙酸乙酯)萃取,接著利用矽膠開放式管柱層析和高效液相層析進行化合物的純化。純物質的化學結構是利用核磁共振光譜、紅外線吸收光譜和高解析電噴游離質譜來進行解析。此外,利用觀察巨噬細胞被刺激後免疫活性因子釋放的生物活性實驗來測試化合物的活性。 最後在本研究中,分離出三種固醇類化合物。其中1和3號化合物具有抑制由脂多醣和三磷酸腺苷所誘導小鼠巨噬細胞中介白素-1β的產生能力,並且抑制活性隨著化合物1和3的濃度有正相關。因此,認為化合物1和3具有發展成免疫調節藥物的潛力。 Mushrooms have been consumed from ancient times, not only for using as nutritionally valuable food but also for the potential as medicine and/or functional food. Recently, much attention has been paid on functional foods due to the increasing of health relative concerns. In previous studies, the bioactive compounds from mushrooms have antimicrobial, antioxidant, antitumor, and immunomodulatory properties. In this research, Morchella esculenta (羊肚菌) was selected for studying. Natural products were extracted by ethyl acetate and purified by silica gel chromatography and HPLC. Chemical structures of purified compounds were identified by using nuclear magnetic resonance (NMR) spectroscopy, fourier transform infrared spectroscopy (FT-IR) and high resolution electrospray ionization mass spectrometry (HR-ESI-MS). Furthermore, bioactive molecules were investigated by biological activity assay, to see the cytokine release after stimulation in mouse macrophages. Three sterols were isolated from Morchella esculenta. Among them, compound 1 and 3 could inhibit the LPS/ATP-induced interleukin-1β (IL-1β) production in mouse macrophage in a dosage-dependent manner. Therefore, the finding suggests that these compounds can have potentials in immunomodulatory medicine. |
