| Abstract: | 本研究是利用螢光光譜法探測幾丁質與幾丁聚醣在不同程度去乙醯化的螢光光譜變化,試著以即時、快速且簡單的方式判斷去乙醯化程度對幾丁聚醣應用上有極大影響,並且進一步尋找結構與螢光的關係。在本研究中,不同去乙醯化幾丁質以紅外光譜判斷去乙醯化程度並配合紫外光譜與螢光光譜觀察其固態下與溶液下吸收光譜與螢光光譜對不同去乙醯的變化。高去乙醯化的幾丁聚醣利用不同含量過硫酸鉀與幾丁聚醣之間的降解反應,探討分子鏈對螢光光譜的關係。以此螢光探測法為基礎,延伸螢光探測上的應用於以幾丁聚醣經蒙脫石改質並以不同程度之戊二醛進行交聯反應所作成的支架,利用螢光光譜法觀察其變化,以期在實際應用面上能以螢光光譜法方便的探測幾丁聚醣結構。 In this study, we used the change of fluorescence parameters ( maximum wavelength , intensity and intensity ratio) of chitosan to correlate with their degree of deacetylation determined by FTIR. Moreover , potassium persulfate (KPS) was employed to degrade the chitosan in acidified aqueous solutions and the results were also correlated with the fluorescence spectroscopy . Furthermore , the fluorescence intensity ratio (550nm/500nm) of the scaffolds prepared by chitosan/montmorillonite nanocomposites were employed to investigate their structure crosslinked by various content of glutaradehyde. |
