Title: 以毛細管區帶電泳法分析幾丁聚醣品質與青脆枝中喜樹鹼含量

Abstract: 以毛細管電泳技術發展出對喜樹鹼(camptothecin)及幾丁質(chitin)之去乙醯率(Degree of Deacetylation, DD)的定量分析方法。前者的分析法乃基於毛細管電泳線上微萃取的技術,可監測在萃取台灣原種青脆枝(Nothapodytes nimmoniana)中喜樹鹼的純化過程。後者乃基於硼酸根離子與胺基之間的錯合反應,此方法可用以估計幾丁物質的去乙醯率,並可應用於檢測食物中的非法添加物-硼砂。
結果顯示,利用DMSO萃取出青脆枝中的喜樹鹼溶液,可以直接注入毛細管電泳儀中做分析,此線上微萃取技術可增加樣品檢測的準確性並減少萃取過程至量測過程所需的時間。由於硼酸根離子與胺基之間的錯合反應改變了泳動率與吸光係數,促進了毛細管電泳的分離與檢測。這兩種分析方法所需的時間均在20分鐘以內。
Capillary electrophoresis procedures for quantifying camptothecin and glucosamine/N-acetyl glucosamine were developed. The former was based on in-capillary micro-extraction technique and can be used in monitoring the purification process of the anti-cancer agent, camptothecin, from a Taiwan medicinal tree, Nothapodytes nimmoniana. The latter was based on the complex reaction between borate ion and the aminosugars; the method is valuable for either determining deacetylation degree of chitinous material or analyzing the illegal food additive, borax.
As the result shown, the DMSO-extract of Nothapodytes nimmoniana can be directly injected, extracted and analyzed with a commercialized capillary electrophoresis system. The borate complex reaction improved the separation and detection by altering the electrophoresis mobility and the optical properties of the aminosugars. Both separation procedures were completed in less than 20 minutes
