Abstract: 光學同調斷層掃描術(optical coherence tomography)在現今之生醫光電領域與醫學研究已成為極重要之三維成像技術。而一般麥克森干涉儀式光學同調斷層掃描系統由於需透過參考臂之前後移動來達成縱向掃描,因此會有額外之震動雜訊、系統體積過大、參考臂光路校準等問題。
本論文中我們展示出一套利用本實驗室自製之雙纖衣結構摻鈰釔鋁石榴石晶體光纖(Ce3+:YAG double cladding crystal fiber)之ASE光源,結合Mirau干涉儀所架設而成之光學同調斷層掃描系統。其具備體積小、極高的空間解析度(橫向7 μm,縱向1 μm)、震動雜訊小等諸多優點。我們利用此系統掃描各種樣本如光柵,液晶面板,單顆細胞等並成功的解析出各種樣本的結構。另外,我們分析除了光源特性之外,聚焦物鏡之孔徑效應對光學同調斷層掃描系統之縱向解析度之影響。
利用本實驗室之Ce3+:YAG OCT系統掃描出之單顆皮膚基質癌細胞(basal cell carcinoma single cell)三維影像,我們藉由相關性(correlation)測試,成功的驗證了單顆細胞OCT影像的可靠性。
Optical coherence tomography (OCT) has become an important scanning technique in the biophotonics realm and medical research. Because the traditional Michaelson based OCT system needs the movement of the reference arm to achieve axial scanning, it will cause additional problems such as vibrational noise, large system volume, alignment of the reference arm, etc.
In this thesis, we demonstrate a Mirau based OCT system using the ASE light source of Ce3+:YAG double cladding crystal fiber and Mirau interferometer. It has many advantages such as compact, ultrahigh spatial resolution, low vibrational noise, etc. We use this system to scan various samples such as grating, liquid crystal panel, single basal cell carcinoma cell and successfully resolve the structure of each sample. And we find besides the property of the light source, the aperture effect of the focusing objective lens will also increase the axial resolution of optical coherence tomography system.
Using the 3D single basal cell carcinoma cell OCT image scanning by our Ce3+:YAG OCT system, we successfully confirm the reliability of single cell OCT image.
