摘要: | 本研究室研發 Monascus purpureus NTU 568 菌株生產之紅麴米,經動物實驗證實,具有預防口腔癌與結腸癌的功效。為進一步鑑定NTU 568 紅麴米中具有預防癌症發生或其他化學保護功效的成份,本研究純化出黃色素 monaphilone A (MA)、monapurpyridine A (MPA)、ankaflavin (AK)、monascin (MS) 與抗氧化成分 deferricorprogen (DFC),再以純物質進行抗癌、抗發炎與抗氧化之研究。利用喉癌與乳癌細胞之細胞模式,證實黃色素 MA、MPA 與 AK 可以活化 caspase-3 與 caspase-9,誘發癌細胞凋亡。利用黃色素 AK 與 MS,搭配大腸癌荷瘤小鼠之動物模式,證實餵食 AK 與 MS 可以抑制腫瘤生成。利用脂多醣刺激小鼠巨噬細胞發炎之細胞模式,證實 MA 與 AK 可以抑制一氧化氮生成、發炎相關酵素與促發炎激素之分泌量。抗發炎動物實驗則使用兩種模式: 利用 12-O-tetradecanoylphorbol-13-acetate (TPA) 誘發小鼠耳朵水腫之模式,證實預先塗抹 MA 與 AK,能減緩體外發炎造成之水腫;利用腹腔注射脂多醣之模式,證實預先投予 MA 與 AK,能降低小鼠血液中一氧化氮生成與促發炎細胞激素之含量。利用橘黴素誘發 HEK-293 氧化損傷之細胞模式,則證實加入DFC,可以顯著降低橘黴素誘發之細胞凋亡。上述結果顯示,含有黃色素MA、MPA、 AK、MS 的 M. purpureus NTU 568 發酵生產之紅麴米,有潛力開發為預防癌症或慢性發炎疾病之機能性食品;而 M. purpureus NTU 568 紅麴米中的抗氧化成分 DFC,則有潛力開發為減緩橘黴素腎毒性之保健成分。 The fermented rice of Monascus purpureus NTU 568 (NTU 568 rice) was proved to possess preventive functions against oral and colon carcinoma. Therefore we executed large-scale preparation from NTU 568 rice to isolate and characterize the cancer preventive or chemo-protective constituents, including four yellow pigments: monaphilone A (MA), monapurpyridine A (MPA), ankaflavin (AK) and monascin (MS); together with a kind of anti-oxidative component named deferricoprogen (DFC). Then we studied on the anti-tumor, anti-inflammatory and anti-oxidative properties of the molecules. First, we employed human laryngeal carcinoma cells and human breast adenocarcinoma cells to demonstrate that MA, MPA and AK could induce some apoptotic events, such as activations of caspase-3 and -9. Then we performed animal experiments and confirmed that feeding AK and MS could inhibit the tumor progressions for colon cancer bearing mice. We also employed murine macrophages cell to demonstrate that MA and AK could suppress the lipopolysaccharides (LPS)-induced inflammatory responses, such as secretion of nitric oxide (NO), inflammatory enzymes and pro-inflammatory cytokines. Moreover, we performed two animal model experiments to evaluate the anti-inflammatory effects of MA and AK. Pre-treatments of these two compounds not only inhibit the TPA-induced ear edema, but also reduced the LPS-induced accumulations of plasma NO and pro-inflammatory cytokines. Finally, we employed human embryonic kidney cells to demonstrate that DFC could minimize the citrinin-induced oxidative damages. All of the findings suggested that the yellow pigment (MA, MPA, AK and MS) in NTU 568 rice could be developed as functional foods against tumor or inflammation-related diseases. We believed that DFC from NTU 568 rice could be developed as a valuable compound against the nephro-toxicity of citrinin. |
