摘要: | 本研究在探討番木瓜花粉生理特性與保存方法,盼有助於番木瓜之育種。利用花粉離體萌芽測試與螢光染色反應檢測番木瓜不同株性、花性、花藥和花朵位置以及不同花齡的花粉活力。試驗結果顯示番木瓜花粉離體萌芽介質以蔗糖10%、硝酸鈣300 ppm、硼酸100 ppm成分組合有較佳的萌芽率。花粉離體萌芽與螢光染色反應檢測結果,番木瓜兩性株雄蕊心皮化花朵的花粉活力均較正常兩性花低。而番木瓜植株性別、花藥位置與花朵著生位置不影響番木瓜花粉活力。番木瓜花粉在花朵初開和盛開時離體萌發率最佳,授粉於盛開雌花的結實數量亦最多。在花開前一週花粉即具有部分稔性,但結實能力較差。而雌蕊的在花開前一週即具有良好的結實能力。部分番木瓜花粉大小畨質性高,有減數分裂畨常的現象,如花粉母細胞發育不同步、形成染色體橋、染色體移動紛亂及小核的形成。以流式細胞儀測定番木瓜花粉的相對DNA含量,得到兩個高峰,番木瓜花粉可能具有2n花粉。2n花粉將有助於應用在番木瓜育種多倍體化的操作。 Studying on the physiological characteristics and preservative methods of papaya pollen will be helpful for papaya breeding. Using FCR (fluorochromatic reaction) and in vitro germination assessed pollen viability of different sex types of tree, floral sex variation, flower and stamen position, and floral stage. The results indicate that the papaya pollen in vitro germination medium consisting of 10 % sucrose, 300 ppm calcium nitrate, 100 ppm boric acid resulted in better germination percentage. The pollen germination percentage and FCR test viability percentage of carpelloid flowers were of papaya pollens lower than these of normal perfect flower (elongate) in hermaphroditic papaya. However, the sex of papaya tree, anther position, and flower position do not affect the viability of papaya pollen. The maximum value of germination was obtained with pollen at the time of full bloom. The highest percentage of pollen germination was found in just anthesis and blooming flowers, pollinating on female flower in bloom also have the largest number of seed. Pollen have viability from one week before anthesis, but seed set was poor. The pistil are well fertilized from one week before anthesis. Some papaya has heterogeneous-sized pollen and meiotic abnormalities, such as the development of pollen mother cells are not synchronized, the formation of chromosome bridges; chromosome movement disturbances and micronuclei formation. Flow cytometric analysis of relative nuclear DNA contents in papaya pollen resulted in two peaks that indicate papaya pollen may have 2n pollen. The 2n pollen can be utilized for manipulating polyploidization in the papaya breeding. |
