標題: 吳郭魚餵食裂褶多醣策略對其非特異性免疫因子之研究

摘要: 本論文研究目的以不同濃度的多醣溶液搭配不同的給予次數觀察對吳郭魚非特異性免疫的影響。利用每天檢測非特異性免疫因子,包含超氧陰離子產生量、一氧化氮產生量及溶菌酶活性,觀察多醣溶液對免疫能力的影響。多醣來源為從裂褶菌培養液中所萃取出的裂褶多醣。
實驗結果發現在實驗一、給予相同次數,不同濃度的多醣溶液情況下,給予一次、二次的實驗組別中以中劑量濃度500ppm及250ppm可以得到較好的免疫能力提升效果,而持續效力也較佳。給予四次實驗組別則以低劑量62.5ppm濃度提升免疫能力效果較佳,持續效力也較好。在實驗二、總劑量相同的多醣溶液,不同分次給予情況下,超氧陰離子產生量在總計量相同情況下,分成二次給予所提升的能力較佳,持續效力較好。一氧化氮產生量在一次給予的情況下提升的能力較佳。溶菌酶活性在分一次、二次給予情況下前期有明顯提升,分四次給予的則在後期有明顯提升。實驗三、相同濃度多醣溶液、給予不同次數的情況下,超氧陰離子產生量及一氧化氮產生量在給予相同劑量多醣溶液的情況下以給予二次組別所提升的效果較佳,持續效力也較長。溶菌酶活性在給予給予高劑量一次或兩次的情況下在前期有提升現象,在給予四次的情況下,在後期有提升的效果。
整體持續效力而言,實驗結果顯示以低劑量62.5ppm給予四次組別最佳,不論是在超氧陰離子產生量、一氧化氮產生量或溶菌酶活性都有明顯提升現象,且與其他組別相比較之下持續效力是較佳的。因此依實驗結果推論,給予吳郭魚β-glucan以增強免疫能力可採用以低劑量62.5ppm給予四次後停止,直到12天後再次給予的餵食策略。
The purpose of this study to know the effect of Schizophyllan is extracted from the culture medium of Schizophyllum Commune on non-specific immune response of Tilapia by oral administration. The preparation different concentrations of polysaccharide solution and with give different number of observations of different Tilapia non-specific immune effects. Daily observe of non-specific detection of immune factors, including the superoxide anion production, nitric oxide production and lysozyme activity.
The results found in (1) to give the same number but different concentrations of polysaccharide solution, given once and twice time experimental group to the concentration of 500ppm and 250ppm in the dose can be better improved results and sustained effect is better. Giving four times experimental groups given low doses of 62.5ppm places concentration was better, sustained results are better. (2) the same total dose of polysaccharide solution, give the times of different circumstances. Superoxide anion production in the medium-dose is given twice into the ability to enhance a better and sustained effect also better than other. Nitric oxide production in the high-dose is given once to enhance the ability to better circumstances. Lysozyme activity in the high-dose and medium-dose given in the previous days has improved significantly, four times given low doses significantly improved in the latter. (3) the same concentration of polysaccharide solution, given the number of different circumstances, superoxide anion production and the nitric oxide production to give twice group are improved to give better results, and the continuing validity longer. Lysozyme activity in the high-dose given once and twice given the circumstances have improved in the early phenomenon, giving four times, in the late enhance the effect.
Continuing validity of the whole, the results show that give the four times groups low doses of 62.5ppm best, both in the superoxide anion production, nitric oxide production and lysozyme activity has improved significantly, and with other groups another comparison, the continued effectiveness is better. Therefore, by inference results given tilapia β-glucan can be used to enhance immunity to low doses of 62.5ppm given four times and stop until 12 days give again the feeding strategy.
