摘要: | 本研究之目的為探討臺灣杉 (Taiwania cryptomerioides) 樹皮精油抑制Hep G2肝癌細胞株生長活性,期盼開發臺灣杉樹皮精油在醫療保健的新用途。以臺灣杉樹皮精油及其成分對Hep G2細胞進行細胞生長抑制評估,發現臺灣杉樹皮精油對Hep G2細胞具有顯著的細胞毒性,樹皮精油的6個分離部中,以S3分離部的細胞毒性最為顯著,S3分離部中的化合物S3A亦同樣具有顯著細胞毒性,效果優於正對照組Etoposide。以流式細胞儀分析經臺灣杉樹皮精油處理後的Hep G2細胞死亡型態,正對照組Etoposide、臺灣杉樹皮精油、S3分離部及化合物S3A處理後的組別,皆可使Hep G2細胞產生細胞凋亡。分析臺灣杉樹皮精油及其成分對Hep G2細胞的細胞週期影響,正對照組Etoposide主要作用於細胞的G2/M期,臺灣杉樹皮精油處理24 hr可輕微抑制Hep G2細胞的G0/G1期,S3分離部及化合物S3A可使Hep G2肝癌細胞G0/G1期的生長停滯。綜合上述結果,顯示臺灣杉樹皮精油及其成分可抑制Hep G2肝癌細胞的生長,具有抗肝癌細胞的潛力。 The aim of this study is to investigate the inhibition of Hep G2 cell (Human hepatocellular carcinoma cell) proliferation by Taiwania cryptomerioides bark oil. Through this study, it is expected that T. cryptomerioides bark oil can be used as pharmaceuticals or healthy products. Bark oil and its compounds are used to evaluate the cytotoxicity of Hep G2 cell. Bark oil has the significant cytotoxicity to Hep G2 cell. S3 fraction is the most effective fraction in cytotoxicity to Hep G2 cell among the 6 fractions. Compound S3A from S3 fraction also significantly induces the cytotoxicity in Hep G2 cell. The cytotoxicity of S3A was even better than that of positive control, etoposide. To confirm the cell death situation, apoptosis analysis of Hep G2 cell was analyzed via flow cytometry. Results showed that etoposide, bark oil, S3 fraction, and S3A induced apoptosis in Hep G2 cell. Effects of bark oil and its compound on the cell cycle distribution of Hep G2 cell were analyzed. Etoposide induced cell cycle arrest in G2/M phase in Hep G2 cell. Bark oil induced cell cycle arrest in G0/G1 phase in Hep G2 cell for 24 hr. S3 fraction and S3A induce cell cycle arrest in G0/G1 phase in Hep G2 cell. According to the above results, T. cryptomerioides bark oil and its compound can inhibit the proliferation of Hep G2 cell with the potential as an anticancer agent against the hepatocellular carcinoma. |
