dc.description.abstract"近年來台灣醫院院內真菌感染的比例逐年上升,其中由白色念珠菌(Candida albicans)所造成的感染佔台灣院內真菌感染比例的一半以上,原因除了罹患後天免疫不全症候群的患者增加,以及因器官移植而使用免疫抑制劑與接受癌症化療等治療導致免疫力低落的病患增加,都使得原先為人體共生菌的白色念珠菌伺機感染人體。 先前實驗室發現到白色念珠菌中ZCF31基因剔除後(Yu-Ting Liao,尚未發表)會導致白色念珠菌zcf31∆突變株的細胞壁增厚,ZCF31 (ORF19.5924)基因為zinc cluster transcription factor (ZCF)之一,在ZCF蛋白質家族中約有80種蛋白質,而目前已知ZCF15與ZCF29和真菌對抗環境中ROS有關,同時也發現ZCF32可以負調控生物膜(biofilm)的形成,但針對於ZCF31的功能還尚未明瞭。 為了瞭解ZCF31基因在白色念珠菌中所扮演的功能,本研究將分為兩個部份做探討,第一部分是檢驗白色念珠菌的致病以及型態轉換能力,了解ZCF31基因的剔除對於念珠菌所產生的影響,針對與白色念珠菌致病力相關的指標:例如附著力(adhesion)、生物膜的生成(biofilm formation)以及opaque-white 轉換等能力進行分析。第二部分則是利用Macrophage Phagocytosis Assay檢驗野生型白色念珠菌SC5314及zcf31∆突變株與巨噬細胞共培養後,巨噬細胞的吞噬情形是否會有差異,進而了解ZCF31的缺失對於白色念珠菌在面對宿主免疫系統會有何影響,最後也透過動物實驗,檢驗在生物體的環境下ZCF31的剔除對於小鼠的存活率所產生的影響。 目前研究結果雖然無法證明基因zcf31刪除後在致病能力上有所改變,但動物實驗顯現出zcf31∆突變株在感染前期似乎具有較高的死亡率,而感染後期zcf31∆突變株的存活率最高,未來希望可以釐清在白色念珠菌中ZCF31基因在致病能力中的功能,同時也了解ZCF31基因剔除後細胞壁增厚的現象對於免疫反應的影響。 "zh_TW
