dc.description.abstract由 1.6 公分長的吳郭魚萃取 mRNA ,加入針對exon l 設計的 degenerate primer 進行 RT-PCR ,得到四種吳郭魚 c-ski cDNA exon l 的核甘酸片段。四種吳郭魚c-ski cDNA exon l 核甘酸片段之中,三種有 309 個核甘酸,一種有 300 個核甘酸。第一種到第四種吳郭魚c-ski cDNA exon l 與雞 c-ski cDNA 第 468 到第 773 核甘酸各有 87.79%、88.35%、 88.35 %以及 88.67 %相同。蛋白質轉譯區由第一個核甘酸到第309個及第300個核甘酸共有103個胺基酸以及100個胺基酸。第一種到第四種吳郭魚 c-ski cDNA exon 1 與雞 c-Ski 從N-端第 101 個胺基酸到第 202 個胺基酸各有 90.10%。 85.15% 8 .15%、82.18%相同。
針對exon l 以及 exon 6 設計 degenerate primer 進行 RT 一 PCR 之後得到第一種以及第二種吳郭魚c-ski cDNA 核甘酸片段,彼此的核甘酸有 76 . 20 %相同。蛋白質轉譯區由第 1 個核甘酸到第1290個以及1452個核甘酸一共有430個以及484 個胺基酸 ,與雞c-ski第 101 個到 599 個胺基酸有54.65%以及56.28%相同。
建立吳郭魚 genomic library ,以吳郭魚 type I c-ski cDNA exon l 核甘酸片段做為探針進行篩選。選殖出一段 genomic DNA 共14 kb ,以 Sacl 作用之後進行南方轉漬分析( Southern blot )。發現吳郭魚 type I
c-ski exon l 位於 4 kb 的核甘酸片段上:其中 exon l 有 894 個核甘酸; 5 ' flanking region 及
5 'untranslated region 有596個核甘酸;其餘為 intron 約有2 . 5 kb 。在 5 ' flanking region 中, ATG 上游-486-502 個核甘酸有 MyoD response element ;-334-341 個核甘酸有 MZF 1 response element ;
-284-293 個核甘酸有 GATA-1 response element ;而 TATA box 則位於 -229-234 個核甘酸的位置上。
