| dc.description.abstract | 黑鯛為一先雄後雌的雌雄同體魚種,在生殖季時可行多次連續或不連續的產卵,其生殖調控機制是由中樞神經系統及內分泌系統所調控,藉由雌二醇控制卵黃生成作用的進行。本實驗以雌二醇處理雄魚,取其mRNA進行反轉錄,經和對照組比較後,發現於21kb處有強烈的差異反應,其極有可能是Vg-mRNA,於是加以分離、純化、切割、大量複製,並加以定序後,得到一條約370bp的cDNA probe。以此探針,用以探知誘發狀態下魚體肝臟內Vg-mRNA/total-RNA的相對比值,由實驗結果發現,處理組的Vg-mRNA/total-RNA比值,於第二次刺激後3hr-6hr到達高峰,且比對照組高過數十倍,實驗的雌二醇處理劑量以lmg/lkg B. W.反應最佳,另外,也以甲狀腺素單獨處理及與二者混合處理,結果發現,單獨處理甲狀腺素有些微的作用,混合處理時則有強大的交互作用,其使高峰更高且往前移,意味著甲狀腺素是一個正向的反應因數。 | zh_TW |
