dc.description.abstract為了對受光蛋白有更深的認識,尤其是彩色受光蛋白(cone pigment)。本實臉用金魚綠色受光蛋白核?酸(goldfish green cone pigment II cDNA)的第552個核?酸到第1364個核甘酸,長約0.8Kb 的DNA作為探針。從鯉魚視網膜cDNA基因庫(cDNA library)中篩選到鯉魚green cone pigment部份核?酸序列及第二種rhodopsin。
完成定序的部份鯉魚putative green cone pigment,核?酸序列長度為772個核?酸。與金魚green cone pigment I,第612到第1151核?酸有94.4%相同;與金魚green cone pigment II,第574到1337個核?酸有90.8%相同。蛋白質轉譯區由第2個核?酸到第482個核?酸,共160個胺基酸。與金魚green cone pigment I,從N-端第190個胺基酸到第349個胺基酸,有95.6%相同:與金魚green cone pigment II,從N-端第190個胺基酸到第349個胺基酸,有93.8%相同。
完成定序的部份鯉魚rhodopsin II,有1253個核?酸;與鯉魚rhodopsin I的第378個核?酸到第1532個核?酸有97.2%相同。在3’端非蛋白質轉譯區,鯉魚rhodopsin II比rhodopsin I多99個核?酸。兩者胺基酸轉譯區有三個胺基酸不一樣。rhodopsin I的第108個胺基酸Ile在rhodopsin II為Val. Rhodopsin I的第169個胺基酸Val在rhodopsin II為 Glu. rhodopsin I的第315個胺基酸His在rhodopsin II為Asn。
我們另外選殖到一段類似人、牛endozepine的cDNA;長度為510個核?酸。5’端非蛋白質轉譯區有42個核?酸。蛋白質轉譯區由第43個核?酸到第303個核?酸,共261個核?酸,轉譯87個胺基酸;與人endozepine胺基酸比較有77.1%相同。3’端非蛋白質轉譯區有207個核?酸。
