dc.description.abstract | 選用rat troponin T exon-15 當 probe 從紅色吳郭魚骨骼肌建立的 cDNA library中,在同一 phage particle中,選殖出二段 cDNA 並加以定序,該 cDNA 片段分別有 2944 及 927 個核甘酸,前者包含5’端末轉譯區的 300 個核甘酸,轉譯區的 1830 個核甘酸,共可譯成 610 個胺基酸和3’端末轉譯區的 615 個核甘酸,此段 cDNA 與 rat TnT 比較,則有 67 %的相似性, T1 轉譯成的胺基酸分子量約 68K daltons ,其 isoelectric point (PI) = 9 . 72 並具有 3 個 N- glycosylation site 及 3 個 cAMP-and cGMP - dependent protein Kinase phosphorylation site .後者包含5’端末轉譯區的 52 個核甘酸,轉譯區的 678 個核甘酸,共可轉譯成 226 個胺基酸及 3 ’端末轉譯區的 196 個核甘酸.而 T2所轉譯成的核甘酸分子量約 26K Daltons ,其 PI 值為 8.24 ,並具有3 個 protein Kinase C phosphsrylation site 。 最後以所得的cDNA insert 作 Northern blot ,分析是否具有組織專一性.在所取得的骨骼肌,平滑肌,心肌,肝,表皮,鰓等 6 個組織中,僅骨骼肌具有正反應,顯示所篩選到的基因為骨骼肌專一性基因。 | zh_TW |
