dc.description.abstract雌性卵生脊椎動物進入生殖週期之初期,在雌二醇誘導下,肝細胞進行卵黃前質合成,經由血液運送至卵巢,以胞飲作用進入卵細胞中,進一步分解為磷卵黃(phosvitin)及脂卵黃( lipovitellin),為堆積於卵細胞中之主要卵黃蛋白,此生理現象為卵細胞成熟之重要特徵。本論文擬就血液雌二醇結合蛋白、卵黃前質鑑定及特性、以及雌二醇誘導卵黃前質生成作用等進行研究,期能針對與卵黃生成作用相關生理獲得更進一步之瞭解。
以 HPLC-陰離子交換管柱層析分析方法,比較雌二醇注射處理性未成熟烏魚血清、雄魚血清與成熟中雌魚血清,鑑定雌性特有、雌二醇誘發之強陰電性蛋白質吸收峰,並比較此吸收峰與卵黃蛋白萃取液之胺基酸組成及免疫性質,確認其為卵黃前質蛋白。以 7% Native-PAGE 配合以各種特性染色分析,確定卵黃前質具有醣蛋白、脂蛋白及磷酸化特性,且其蛋白質相當不穩定。經由 10% SDS-PAGE 分析,可知卵黃前質蛋白主要由五條太鏈所組成,其大小均接近於 116kd。
以不同劑量之雌二醇注射處理性未成熟魚,約可誘導卵黃前質生成;,則血清中卵黃前質最高含量之升高與注射劑量呈相關性。以 12.5 ug/g 體重雌二醇劑量注射尚未進入生殖季之成魚,其血清中之卵黃前質含量在第十天可達 121.73 士 33.73 mg/ml,遠高於生殖季內成熟烏魚採樣測值(0-2.6 mg/ml), 其可能原因為卵黃前質在非生殖季中無法進入卵細胞,而造成高量蓄積於血液中,或因雌二醇注射劑量高於一般之生理值,而造成之藥理現象。
烏魚血清中,證實有與雌二醇具專一性結合能力蛋白之存在。以不同之類固醇進行競爭性結合,發現其與一般性類固醇結合力高,但與助孕素之結合能力不佳;經以 HPLC-陰離子交換管柱分離雌二醇結合蛋白並進行 7 % Native-PAGE 分析,發現雌二醇放射性集中於 Rf 值 0.43 處,但可能因蛋白質含量過低,而未能對應於一電泳染色帶。
