dc.description.abstract | 梭德氏蛙(Rana sauteri)是臺灣產特有種兩生類之一,不但廣泛分佈於全島的山區,而且可從低海拔(0-500公尺)起至海拔三千多公尺均可生存。由於梭德氏蛙的生殖季節隨地點而異,有回到相同的繁殖場所生殖之記錄,而且在不同的海拔高度的成蛙之呼吸代謝作用亦有不同,故不同族群間的基因組成可能因長期的生殖隔離與缺乏基因流動(gene flow)而有所變異。關於梭德氏蛙的研究中,目前已有生態、生理及細胞遺傳等方面的資料,若能增加族群遺傳方面的研究,則更完整。 本研究利用聚合?鏈鎖反應(PCR)的兩股定序法(double-strand sequencing)來分析分屬於10個族群的63段梭德氏蛙的粒線體DNA序列,嘗試瞭解在D-loop區域輕股的5’端附近之基因組成。而研究結果顯示:梭德氏蛙的粒線體DNA的變異包括鹼基替代(base substitution)、長度變異(length variation)及基因重排(gene rearrangement)。在粒線體DNA的控制區域(control region)有一段由39個鹼基組成,重覆十次以上的直接重覆片段(direct tandem repeat)。另外,發現有些樣本檢測出具有不同的mtDNA型態(異質性,heteroplasmy)。 為瞭解梭德氏蛙在不同族群的地理變異和親緣關係,將所分析的63段DNA片段可歸納出18種基因型,主要區分成屬於濁水溪水系的中部系統和淡水河水系的北部系統,而且各水系內的不同族群有不同的基因型組成,顯示出各族群之間的地理親緣關係可用水系作為主要的區隔。本研究顯示粒線體DNA之D-loop區域是探討無尾兩棲類的族群遺傳及演化之良好工具。 | zh_TW |
