dc.description.abstract魚類卵細胞成熟過程中,當達到卵黃生成期(Vitellogenesis ) 後,即暫時停止發育,迄受到適當環境因數刺激,經由神經及內分泌調控,腦下垂體分泌促性腺激素(Gonadotropin , GtH)進而誘發類固醇生成作用(Steroidogenesis) ,並合成成熟誘導類固醇 (Maturation-Inducing Steroid , MIS ) ,即17α,20β-Dihydroxyprogesterone , 待卵細胞質內之成熟促進因數(Maturation -Promoting Factor , MPF )被活化後,促使卵細胞進入最後成熟(Final maturation)。大多數魚種之卵細胞在最後成熟期間會有吸水(Hydration)的現象,此乃卵細胞滲透壓調節之結果。本研究以烏魚(Mugil cephalus)為材料,利用人類絨毛促性腺激素(Human chorionic gonadotropin , hCG)催熟,使其進入最後成熟,並依卵細胞型態特徵及卵核位置,將卵細胞分為卵核居中期( Subperipheral stage , S )、卵核移動期(Germinal vesicle migration , GVM )、卵核破裂期(Germinal vesicle breakdown , GVBD)、排卵期(Ovulation , OV )等四期,並以成熟卵細胞(即卵黃第三期,Tertiary globule stage , YG3)為對照組,測定最後成熟各階段中卵細胞內與血清中滲透壓的改變,同時並測量血清、卵細胞中Na+、K+、Ca+2、Mg+2及Cl- 五種離子之含量變化及卵中有機物、灰分、卵徑及水分含量,以瞭解吸水過程中,卵細胞生化組成之改變。結果發現:卵徑與水分含量隨最後成熟的進行,呈正相關的增加,而卵細胞滲透壓及其總陽離子含量亦有相同關係。此結果顯示,最後成熟期間卵細胞體積的增大為吸水之結果,而吸水現象的發生,乃卵細胞內大量離子進入以達到滲透壓調節所造成。而卵細胞中K+的大量增加,可能表示對烏魚而言,K+為影響吸水之重要因數。本研究亦測定最後成熟各階段的血清中,17β-estradiol (E2)及17α-hydroxyprogesterone (17α-OHP ) 之濃度變化。結果發現:二者之濃度皆隨最後成熟之進行而減少。而17α-hydroxyprogesterone 乃多種成熟誘導類固醇之前驅物,其濃度之降低,表示著成熟誘導類固醇分泌量之增加,以促動最後成熟的進行。zh_TW
