dc.description.abstract利用顯微微量注射或電穿孔法,將外源性異質基因導入受精卵內是一般常用的基因轉殖魚體的方式。本研究係探討電穿孔法促使精子攜帶外源性生長激素異質基因及成長觀察,所使用外源性生長激素異質基因是鮭魚的生長激素 cDNA ( opAFP GHcDNA ) (以抗凍蛋白的?動子為主控),藉著電穿孔處理將此基因導入泥鰍(Misgurnus anguillicaudatus)的精子後用於人工受精,所得的受精卵 70740 士 270 個、受精率 79 %、孵化率 61 % ,此與沒有處理的控制組相比,在受精率有顯著的差別 ( < 0 . 05 )、但在孵化率上差別不顯著。以放射性標識的 opAFP GHcDNA 片段為探針,經點墨法( dot blot )偵測180 尾 15 天的魚苗其 genomic DNA 有 94 尾呈正反應,再經南方氏點墨法 ( Southern blot )在 4.0kb 的位置有正反應,以 PCR 的方法偵測 8 尾魚齡 240 天的成魚有 7尾呈正反應、但呈鑲嵌現象,再以三組 primers 經 PCR 偵測確定 opAFP GHcDNA 的存在。經過 6 個月的成長觀察,第二個月起實驗組產生各式各樣的畸型,但其平均成長率( growth rate )的比較,實驗組為控制組的1.3倍,這個實驗證明瞭以電穿孔法促使泥鰍的精子作為傳遞外源性異質基因的媒介是可行的。zh_TW
