dc.description.abstract本論文之目的在於探討人類絨毛膜促性腺激素(Human Chorionic Gona dotropin , HCG )和前列腺素F2α ( Prostaglandin F2α PGF2α)參與香魚( Plecoglossus altivelis )卵細胞最後成熟作用之程度,以瞭解其基本的生理作用途徑。本研究選取卵黃生成後期( Postvitellogenesis )和最後成熟期(Final maturation)之卵細胞,依照性腺成熟指數(Gonadosomatic index , GSI )、卵徑大小、卵核位置及濾泡層之形態,將其區分為六個成熟期,分別為卵黃第三期( Tertiary yolk globule stage , YG3 ) ,卵核居中期( Subperipheral stage , S ) ,卵核移動前期( The early stage of germinal uesicle migration , GVMI ) ,卵核移動後期( The late stage of germinal vesicle migration )、卵核破裂期( Germinal vesicle breakdown stage , GVBD )及排卵期( Ovulation , O )。以人類絨毛膜促性腺激素刺激素、前列腺素 F2α 單獨或協同刺激各成熟期之卵細胞,得到各成熟期卵細胞對人類絨毛膜促性腺激素和前列腺素F2α 之反應程度,其中以卵核居中期卵細胞受人類絨毛膜促性激素作用程度最大,隨著卵細胞的成熟,人類絨毛膜促性腺激素對卵細胞之作用程度漸減,至卵核破裂期略有恢復,但隨著排卵作用,其對卵細胞之作用程度又再度下降。卵細胞從卵黃第三期到卵核移動末期,人類絨毛膜促性腺激素刺激卵細胞之最高反應程度,除卵核移動前期為 1 IU HCG/ml 外,皆為 5 IU HCG / ml ,而前列腺素F2α 。在卵核破裂期卵細胞協助 5 IU HCG / ml 刺激卵細胞之最低有效劑量介於 2 -4 μ M / ml 之間.對於加入β型腎上腺素接受器之促進劑-isopoterenol 和抑制劑-propranonol 協同人類絨毛膜促性腺激素刺激卵細胞之作,其結果顯示, isopoterenol 和 propranolol對 HCG 刺激卵細胞之作用程度,確有促進和抑制之作用,特別是在卵核移動前期之卵細胞,其促進或抑制 HCG 對卵核移動期卵細胞之刺激作用的最低有效濃度,在 isoproterenol 為 0.5 - 4μg/ml之間,而在 Propranolol 則高於 20μg/ml 之濃度,由此結果可知,促性腺激素接受器之作用機制與β型腎上腺素接受器十分相似,不僅都是 Gs 蛋白伴隨的接受器(G-protein coupling recepor ) ,並以活化腺核?酸酵素,產生 cyclic AMP 為其二級訊息傳遞者,且在抑制和促進接受器活性上有其共通處,值得更深入探討。zh_TW
