dc.description.abstract | 中文摘要 香魚之卵黃前質經由電泳分析、西方點墨法、雌性素誘導、胺基酸組成分析及染色法,確定其為雌魚所特有的磷酸醣脂蛋白,與卵黃蛋白具有免疫交叉反應,而且雌性素可誘導其生成。在自然膠體電泳中,卵黃前質含有三分子,分子量分別為 552kd , 291.5kd 及 270kd ,其關係可能為 Dimer 及 Monomer ;在 SDS-PAGE 分析,則具有 12 條勝?鏈,所以卵黃前質為複雜的大分子。 以單一步驟之高效陰離子交換層析法,可自血清中純化出高純度且高濃度之卵黃前質。由火箭式免疫電泳作定量分析,得知香魚於卵巢發育成熟過程。血清中卵黃前質之濃度變化由發育初期的0.1mg /ml ,逐漸增加卵巢最後成熟之 15 mg /ml ,以此方法定量,靈敏度可達 70ng,僅需 2ul 稀釋後之雌魚血清。此外,血清中之雌二醇含量,可由高壓逆相層析法分離且定量,靈敏度約 100 pg ,其濃度分佈範圍約 5-15ng /ml 。比較雌、雄魚肝細胞之電顯切片,發現雌魚肝細胞之粗糙內質網。高基氏體非常發達,顯示其蛋白質生合成作用非常旺盛。而雄魚經雌二醇注射後,亦有相同的結果。 此外,卵黃前質及卵黃蛋白之免疫特性,及其他生理變化,亦一併探討之。 | zh_TW |
