| dc.description.abstract | 本實驗以鰻(Anguilla japonica)之腎臟、生殖腺（卵巢和精巢）做EK，EO和ET細胞的初級培養。利用EK細胞的懸浮培養，比較培養溫度，培養液中血?含量，NaCl濃度和pH?，以及初植細胞密度等各項因素對細胞培養之影嚮，以尋求最適合的細胞培養條件。依實驗結果知培養液?Eagle's Minimum Essential Medium和Tissue Culture TC 199二者以一比一的混合液，添加10％ fetal calf serum, NaCl濃度?0.137M, pH??7.2－7.5，初植細胞密度?1.0×105 cells/ml-106cells/ml，培養溫度?25℃等之培養條件。 腎臟組織因?淋巴球很多，纖維很少，不容易培養出monolayer。但EO和ET細胞在上述最適當的培養狀況下，於培養後3—7天，皆可長成monolayer。又不拘EO或ET組織，長成monolayer之細胞皆?織維樣細胞和上皮樣細胞。用消化酵素0.1％ trypsi與Versene之等量混合液分離細胞可進行斷代培養至第三代。 | zh_TW |
