dc.description.abstract本實驗以鰻(Anguilla japonica)之腎臟、生殖腺(卵巢和精巢)做EK,EO和ET細胞的初級培養。利用EK細胞的懸浮培養,比較培養溫度,培養液中血?含量,NaCl濃度和pH?,以及初植細胞密度等各項因素對細胞培養之影嚮,以尋求最適合的細胞培養條件。依實驗結果知培養液?Eagle's Minimum Essential Medium和Tissue Culture TC 199二者以一比一的混合液,添加10% fetal calf serum, NaCl濃度?0.137M, pH??7.2-7.5,初植細胞密度?1.0×105 cells/ml-106cells/ml,培養溫度?25℃等之培養條件。
腎臟組織因?淋巴球很多,纖維很少,不容易培養出monolayer。但EO和ET細胞在上述最適當的培養狀況下,於培養後3—7天,皆可長成monolayer。又不拘EO或ET組織,長成monolayer之細胞皆?織維樣細胞和上皮樣細胞。用消化酵素0.1% trypsi與Versene之等量混合液分離細胞可進行斷代培養至第三代。
