dc.description.abstract為欲瞭解鯉魚卵細胞發育過程中之基因表現情形。本論文首先建立了卵原細胞期與高級卵黃期之鯉魚卵巢cDNA基因庫。經由兩基因庫兩萬重組基因株之差異篩選(differential hybridization)找出表現不同的cDNA約20個。將其中五個cDNA( Cov7, 10, 23, 24, 33)轉入質體,然後再以之為探針,對不同發育時期之鯉魚卵巢進行RNA dot雜交反應,結果發現Cov7, 23, 24與33等四株基因在發育早期卵細胞有明顯的表現,而Cov10基因則在卵細胞發育的過程中均有明顯但不等程度的表現。將核酸定序結果在“DNA Star”基因庫中比對。結果發現0.5kb長之Cov10為ubiquitin基因,但1.4kb長之Cov7則尚不知為何基因。zh_TW
dc.description.abstractIn order to study the gene expression during oogenesis in carp, two λgt10 cDNA libraries were constructed from carp ovaries of oogenium and tertiary yolk stages, respectively. Through differential hybridization of twenty thousand recombinant phages, twenty cDNA clones were obtained. Five of them, Cov7,10,23,24 and 33, were subcloned into plasmids and used as probes to study their expression during oogenesis. RNA dot analysis indicates that Cov7,23,24,33 are slightly expressed in the oocyte of young developmental stage and Cov10 is highly but differentially expressed throughout the oogenesis. After computer search of nucleotide sequences in the “DNA Star” gene bank, the 0.5 kb Cov10 is the gene encoding ubiquitin and a fuse protein. None of the reported genes is corresponding to the 1.4 kb Cov7.en
