本實驗的目的是將幾種動物之腦下腺性腺刺激素(Gonadotropin, GTH)的次單元己分別加以重組,然後測定這些重組分子促進睪九產生雄性素的能力,以期瞭解各次單元的特性及其在生物活性上所扮演的角色。
鯉科的鯉魚、白鰱和大頭鰱GTH之次單元可以任意置換,而不影響促進離體鯉魚睪丸組織產生雄性素的能力,顯示鯉科中此三種魚類GTH之次單元的屬性非常的類似。
在海鰻與大頭鰱GTH次單元的重組分子中,海鰻GTHα次單元與大頭鰱GTHβ次單元之重組分子及大頭鰱GTHα次單元與海鰻GTHβ次單元之重組分子均具有促進離體鯉魚睪丸組識產生雄性素之能力,但前者之活性較後者為佳。
鯉魚GTH與羊黃體成長激素(Luteinizing hormone, LH)次單元的重組實驗中發現,除了鯉魚GTH的α,β次單元之重組分子,對離體鯉魚睪丸生生成雄性素具有顯著之促進作用外,羊LHα次單元與鯉魚GTHβ次單元的重組分子以及鯉魚GTHα次單元與羊LHβ次單元重組分子也具有促進離體鯉魚睪丸組識生成雄性素之能力,但其活性較弱,分別為鯉魚GTHα及β次單元之結合分子活性的60.3%及29.5%,至於羊LHα次單元與β次單元之結合分子則全然無活性。而在老鼠萊氏細胞生成雄性素之測定系統中,若以羊LH的α次單元與β次單元的結合分子的活性為100%。則鯉魚GTHα次單元與羊LHβ次單元結合分子的活性為10.0%,而羊LHα次單元與鯉魚GTHβ次單元的結合分子則幾無活性。
綜合本實驗利用不同種動物的GTHα次單元及β次單元進行重組之結果顯示,α次單元則在不同種類間仍具有相似性,故可彼此互換而形成具有生物活性的雜種GTH分子,而β次單元,則有明顯的種別性差異,故可決定激素的屬性。此等事實則顯示,脊椎動物的GTH在演化過程中,α次單元較為保守,但β次單元則顯得分歧。
