dc.description.abstract | 將兩種單倍體野生菸草Nicotiana plumbaginifolia與N. sylvestris的葉肉原生質體以PEG誘導融合,在無人?篩選的情況下培養使長成植株。檢查早期分化出植株的259個癒合組織之染色體,發現98.8%均?雜種。將118株雜種(原自32個癒合組織)之葉綠體DNA以Hind Ⅲ限制?切割,由限制?圖譜分析其葉綠體來源,發現除兩株雜種含雙親的葉綠體外,其餘均僅含單一親代的葉綠體。有的癒合組織分化出來的植株均含相同單親的葉綠體,有的癒合組織分化出來的植株含不同親代的葉綠體。在SP,SSPP,SSP及SPP 4種染色體組組合的雜種中,含N. sylvestris的葉綠體與含N. plumbaginifolia的葉綠體的植株數,均符合1:1,顯示在體細胞雜種中葉綠體之分離是逢機的。 | zh_TW |
dc.description.abstract | Mesophyll protoplasts of haploid Nicotiana plumbaginifolia and N. sylvestris were fused by polyethylene glycol induction. Plants were regenerated from 259 calli without any selection pressure and characterized by cytological examination. Most of the calli (98.8%) differentiated into hybrid plants. Chloroplast DNAs of 118 somatic hybrid plants regenerated from 32 calli were digested with Hind III and the resulting patterns were analyzed. The results revealed that the hybrids contained chloroplasts of one or the other parent; however, 2 hybrids contained chloroplasts of both parents. Some calli regenerated hybrid plants having identical chloroplasts, while others differentiated plants having different kinds of chloroplasts. The assortment of chloroplasts was random in the somatic hybrids regardless the ratio of the two parental genomes. | en |
