dc.description.abstract本論文分為兩部分:
第一部分利用光學顯微鏡術和電子顯微鏡術觀察泥鰍表皮中四種主要細胞:基底微絲細胞,微絲細胞、棍狀細胞、黏液細胞形態及表皮組織發生過程中細胞的分化和生長。另外,淋巴球在泥鰍表皮發生過程中的源起和分佈在本文中亦有初步的瞭解。
第二部分係利用注射氚-胸腺核?的自動放射顯影術,探討泥鰍表皮細胞的分裂和更生現象,結果顯示:
1.泥鰍表皮細胞的分裂並不像哺乳類表皮細胞的分裂只限於底層,而在表皮上、下層都有細胞分裂的現象。
2.棍狀細胞與黏液細胞不具細胞分裂能力,應為極度分化的細胞。
3.微絲細胞的分裂頻度甚高,部分細胞可能扮演著始細胞(progenitor cell)的角色,棍狀細胞和黏液細胞可能來自此類細胞的分化。
4.部分基底微絲細胞可能扮演著幹細胞(stem cell)的角色。這種細胞位於表皮底層,染色較深,與其他微絲細胞有明顯區別。細胞內含有二種不同型態的微絲構造,分別位於細胞質底部和頂部。當此類細胞進行分裂,紡綞體方位與基底膜垂直時,兩個子細胞會分別分配到兩種不同微絲構造,上側的子細胞所獲得的微絲與表皮其他微絲細胞所含微絲型態一致,極可能往上移動而扮演上述始細胞的角色。下側的子細胞則繼續留在底部,扮演幹細胞的角色,這樣一種不對稱的細胞分裂,正是一個幹細胞所需具備的特性之一。

dc.description.abstractThe histogenesis of the loach epidermis was examined with light and electron microscopies. Morphological features and the development of the four main cell types, namely basal filament-containing, filament—containing, club and mucous cells are presented.
Origin and distribution of the other cell type-lymphocyte in the histogenesis of loach epidermis are also discussed.
Cell renewal of the loach epidermis was revealed by autoradiography with the injection of Tritated Thymidine. The results indicate that:
1. Cell divisions occur not only at basal region but also at superficial region of loach epidermis, unlike the epidermis of higher vertebrates which are characterized with a germinal basal layer.
2. The club and mucous cells are highly differentiated, and would not be able to divide mitotically.
3. Most of the filament-containing cells might serve as progenitor cells. They divide actively and may differentiated into club and mucous cells.
4. The basal filament-containing cell could be the stem cell. The basal FC cell rested on the basement membrane of the epidermis, are stained distinctly from other FC cells, contain two types of filament localized within basal and apical cytoplasm respectively. When the mitotic spindle is accord with the baso-apical axis, the cell division will lead to the segregation of the apical and basal cytoplasm. This unsymmetrical division would provide mean to generate two different daughter cells. The distal cell may serve as proginator of the filament-containing cell in general, and the basal cell remains associated with the basement membrane is saved as the stem cell.
