| dc.description.abstract | 本實驗利用添加20%-10%胎牛血清的L-15培養基建立了花身雞魚(Therapon jarbua FORSK?L)胸鰭來源的永久性細胞株(JF),並針對此細胞株做各種特性的研究。 此株化細胞經顯微,超微及染色等觀察顯示,細胞形態兼具纖維狀及表皮狀。細胞在基質上貼平鋪展,細胞表面平滑,鮮有微絨毛等突起。細胞間有絲狀或葉狀偽足接觸。細胞內,細胞核呈不規則形,粒線體,內質網及高氏體均很發達,柱狀的粒線體以近核區為中心散射,內質網內含有許多合成物質,顯示細胞的代謝功能或分泌功能都很旺盛。各種液態培養環境中,JF細胞株最適合的環境下,其細胞數目倍增的時間約2天。pH 6-8或滲透壓200 mmol/Kg-650 mmol/Kg的條件下細胞均可生長。以pH 6.5-7.5和滲透壓450 mmol/Kg左右的環境下最適合生長。不同的pH,滲透壓,血清濃度和培養溫度對細胞的生長速率,附著效率都有影響。細胞的大小會隨pH增高而增大,在高溫(37℃)培養時細胞也會變大。pH及參透壓過高或過低均會造成細胞病變。染色體數目的分佈從10幾條至90幾條都有而具48條的細胞最多。群落形成率在1.7%以下。 由實驗結果顯示此株化細胞具停泊依賴性,血清依賴性密度,依賴性等正常細胞的特性。對於Infectious Pancreas Necrosis Virus (IPNV) 病毒AB, VR-299, Sp等株及CSV病毒均有感受性,平均每個細胞的病毒產量IPNV約420-1327 TCIDoo/cell, CSV約9 TCIDoo/cell。細胞在液態氮中貯存8個月仍有95%的回收率,且尚未被細菌、黴菌、黴漿菌或病毒污染。本細胞株適合細胞生物學,病毒學或其他方面的研究。 | zh_TW |
