本實驗的目的在於分離草魚腦下腺腎上腺皮質刺激素(Adrenocorticotropin, ACTH)。
草魚腦下腺以0.1N醋酸萃取,將上層液冷凍乾燥,利用HPLC做初次分離,分別以階梯式滌流,分別收取10%、40%、60%、80%之氰甲烷(含0.07% TFA)滌流物,其中10?40%氰甲烷(含0.07% TFA)滌流物具最強之皮質刺激素活性。將此部分冷凍乾燥後,再以HPLC做第二次分離。
第二次分離時,以10?40%氰甲烷(含0.07% TFA)展開,滌流1.5小時,使最終氰甲烷濃度達於40%。每5分鐘收取一分液,冷凍乾燥後做生物活性測定,其中以F7、F8、F9、F10之活性最強,經劑量效應,以4倍及16倍稀釋,再測生物活性以及測F7?F10之間各吸收?之活性,發現以F8、F9具最強的皮質刺激素活性。分別收集此二部分,匯在一起,冷凍乾燥後以HPLC做第三次分離。
第三次分離時,同樣以第二次HPLC解析條件解析之。將各吸收?回收做生物活性測定,結果得一活性極強且單一之吸收?的物質。
