Title: 鴨腦下腺生長激素蛋白質及基因的研究
The Study of Duck Growth Hormone: Protein and its Gene

Abstract: 利用HPLC來純化鴨腦下腺生長激素,以此純化所得之鴨生長激素,決定了其N端的胺基酸序列,發現鴨與雞生長激素的N端幾乎相同。
將鴨腦下腺的polyA(+)mRNAS純化後當作範本,來合成雙股cDNAs,經acrylamide gel電泳純化過後,建立鴨腦下腺cDNA library。以p32-標識豬生長激素cDNA為Probe,從其中篩選出鴨生長激素cDNA clone,經DNA定序後,我們知道鴨生長激素及其先驅物的一級結構。
以鴨生長激素cDNA植入表現載體,在大腸菌內表現,以放射免疫定量法初步測得少量表現。
The growth hormone was purified from duck pituitary glands. Using this purified duck growth hormone, we have determined its N-terminal 20 amino acid sequence, which is virtually identical to that of chicken growth hormone.
Messenger RNAs isolated from duck pituitary glands were used to construct a duck pituitary cDNA library. A clone containing duck growth hormone cDNA was isolated by using 32P-labelled porcine growth hormone cDNA as probe and sequenced. The amino acid sequence of duck growth hormone precursor has been deduced from the DNA sequence.
The duck growth hormone cDNA was used to generate a vector capable of producing duck growth hormone in E.coli. A polypeptide was produced in E.coli with the predicted size, but the amount of expression was low by radioimmunoassay.
